摘　　要：目的检测Beclin1、PTEN在鼻咽癌组织中的表达水平,分析自噬蛋白Beclin1与凋亡相关蛋白PTEN在鼻咽癌发生发展中的作用及临床意义。方法应用免疫组织化学方法检测50例鼻咽癌、20例癌旁黏膜和15例鼻咽黏膜慢性炎组织中Beclin1和PTEN蛋白表达,并分析其与临床分期、病理组织类型和转移的关系。结果Beclin1在鼻咽癌、癌旁黏膜和鼻咽黏膜慢性炎组织中阳性表达率分别为38.0%、65.0%、93.3%,PTEN的阳性表达率分别为34.0%、85.0%、93.3%,Beclin1和PTEN蛋白表达程度都与鼻咽癌临床分期、组织学分型和颈部淋巴结转移相关(P<0.05),且Beclin1与PTEN在鼻咽癌中的表达呈正相关(rs=0.946,P<0.05)。结论鼻咽癌组织中Beclin1、PTEN低表达与鼻咽癌发生、发展相关,检测其表达对研究鼻咽癌发生、发展有重要的作用,并可作为患者预后判断的重要标志物。Objective To investigate the expression of Beclinl and PTEN protein in nasopharyngeal carcinoma（NPC） and to evaluate their clinical implications in carcinoma pathogenesis, development, or metastasis of NPC. Methods Immunohistochemical method was used to determine the expression of Beclinl and PTEN protein in 50 NPC cases, 20 cases of nearby mucosa of the carcinoma and 15 cases of chronic inflammation of nasopharyngeal mucosa. The relationship between the gene expression and clinicopathological features, metastasis was analyzed. Results The positive expression rates of Beclinl in NPC, nearby mucosa of the carcinoma and chronic inflammation of nasopharyngeal mucosa were 38.0%, 65.0% and 93.3%, zrespectively, and PTEN were 34.0%,85.0% and 93.3%. The expression of Beclinl and PTEN were positively correlated with the clinicopathological features, tissue types and neck lymph node metastasis in NPC（P〈0.05）. The expression of Beclinl in NPC was positively related to the expression of PTEN（rs = 0.946, P〈0.05）. Conclusion Reduced or absent expression of Beclinl and PTEN were observed in NPC tissues and they were associated with tumor occurrence, progression and metastasis of NPC. These 2 proteins are important indicators in predicting the progression and prognosis of patients.
