摘  要:目的观察核转录因子KB(NF-kB)拮抗剂对高血压大鼠下丘脑室旁核中血管紧张素Ⅱ受体(AT1-R)水平的影响,探讨脑内NF-kB是否通过调节下丘脑室旁核(PVN)中的AT-R在高血压发病中发挥作用。方法选取体重250-275g雄性SD大鼠,随机分为4组:高血压干预组(ANGⅡ+PDTC)、高血压对照组(ANGⅡ+VEH)、假手术干预组(NS+PDTC)和假手术对照组(NS+VEH)。记录3d基础血压后,麻醉状态下肩胛区皮下给予ANGⅡ(200ng·kg^-1·min^-1)诱导高血压,对照组给予生理盐水(Ns)。干预组通过下丘脑室旁核插管给予吡咯烷二硫基甲酸盐(PDTC);对照组给予人工脑脊液(VEH)。术后采用无创血压测量系统监测血压,4周后使用免疫荧光技术检测下丘脑室旁核AT1-R水平。结果ANGⅡ+VEH与NS+VEH相比,下丘脑室旁核中AT1-R水平明显升高,血压升高(P〈0.05);ANGⅡ+PDTC与ANGⅡ+VEH相比,下丘脑室旁核中AT1-R水平降低,血压下降(P〈0.05)。结论中枢NF-kB通过调节下丘脑室旁核AT1-R水平参与高血压的发病机制,阻断NF-kB可在一定程度上降低血压值。Objective To investigate the effects of NF-KB blockade in the paraventricular nucleus of hy- pothalamus (PVN) on the level of ATj-R in hypertensive rat, and to explore the possible role of brain NF-KB in hypertensive rats through modulation of AT1-R in paraventricular nucleus (PVN). Methods Adult male Sprage- Dawley rats(250-275 g) were randomly divided into 4 groups, hypertension treatment group(ANG Ⅱ +PDTC ), hy- pertensiofi control group(ANG Ⅱ +VEH), sham treatment group(NS+PDTC), sham control group (NS+VEH). Af- ter three days' baseline blood pressure recordings, rats were implanted subcutaneously with an angiotensin II 200 ng· kg-l ·min-1 filled osmotic minipump to induce hypertension. Rats in sham group were administered with saline (NS). PVN cannulae were implanted for the injection of PDTC (5 μg/h) and artificial cerebrospinal fluid (VEH, 0.25 μ1/h). Immunofluorescence was used to test the level of AT1-R in PVN 4 weeks later. Results Hy- pertensive rats showed a significant increase in the expression of AT1-R in the PVN when compared with SHAM rats (P〈0.05). Treatment with PDTC was shown to reduce the AT1-R level (P〈0.05). Conclusion Brain NF- KB involves in hypertension by modulating AT1-R in the PVN, blocking NF-KB can reduce blood pressure.
