香烟烟雾暴露对大鼠肺组织血小板衍生生长因子BB表达的影响

摘  要:目的研究烟雾暴露对大鼠肺组织血小板衍生生长因子BB(plateletderivedgrowthfactor-BB,PDGF-BB)和核因子eB(nuclearfactor-kB,NF—kB)p65表达的影响,探讨烟雾暴露致大鼠肺动脉高压的发生机制。方法雄性Wistar大鼠32只,随机分为对照组、烟雾暴露4、8、12周组,每组8只,制备各组烟雾暴露致肺动脉高压模型。RT-PCR方法检测PDGF-BBmRNA表达,免疫组织化学方法测各组大鼠肺组织PDGF-BB、NF-kBp65蛋白表达。结果①大鼠肺组织中PDGF-BBmRNA相对表达量在烟雾暴露4周组(2.823±0.453)、8周组(4.015±0.052)、12周组(6.098±0.439)高于对照组(1.024±0.231),差异有统计学意义(P〈O.05);②大鼠肺组织中PDGF-BB蛋白相对表达量在烟雾暴露4周组(O.220±0.005)、8周组(o.367±0.004)、12周组(O.492±0.003)高于对照组(0.077±0.003),差异有统计学意义(P〈O.05),NF-kBp65蛋白相对表达量在烟雾暴露4周组(0.330±0.003)、8周组(o.452±0.003)、12周组(O.596±0.003)高于对照组(0.186±0.001),差异有统计学意义(P〈O.05);③PDGF—BB蛋白的表达与PDGF-BBmRNA的相对表达量及与NF-kBp65蛋白的表达均呈正相关(r=0.686、r=0.501,P值均〈O.01)。结论烟雾暴露大鼠肺组织中PDGF-BB在转录和翻译水平表达均增高,且PDGF-BB和NF-kB蛋白表达有相关性,提示PDGF-BB和NF-kB在烟雾暴露引起肺动脉平均压增高的过程中可能发挥了一定作用。Objective To study the mechanisms of pulmonary hypertension caused by smoke exposure in researching the effect of smoke exposure on the expressional levels of platelet-derived growth factor-BB (PDGF-BB) and nuclear factor-kB(NF-kB)p65 in rats lungs. Methods 32 male Wistar rats were divided randomly into control group, smoke exposure 4,8,12 weeks group. The rat models of all groups were established by smoke exposure. The relative expression of PDGF-BB mRNA was detected by RT-PCR and the relative expression of PDGF-BB and NF-kBp65 protein in rat lungs were detected by immunohistochemistry in all rats of four groups. Results (1)The relative expression of PDGF-BB mRNA of rats in smoke exposure 4 and 8 and 12 weeks groups were significantly higher than the control group (all P 〈0.05). QThe expression of PDGF-BB protein in the lung tissues and NF-kBp65 protein were significantly higher than those of the control group (all P d0.05). (2)The expressions of PDGF-BB protein were positively correlated with the relative expression of PDGF BB mRNA and the NF-kBp65 protein. Conclusions Smoke exposure can increase the expression level of PDGF-BB not only on the transcription but also the translation level, PDGF-BB and NF-kB play an important role in pathogenesis of pulmonary hypertension caused by smoke exposure.
