摘 要:目的利用高通量筛选方法进行刺糖多孢菌诱变选育获得多杀菌素高产菌株。方法以ASAGF73为出发菌株,通过亚硝基胍(NTG)诱变,并利用96孔板发酵培养结合快速生物测定进行高通量筛选。结果诱变剂量为2mg/mL,诱变时间50min时突变株多杀菌素产量有较大幅度提高。通过3轮复筛验证最终获得8株产量分别比出发菌株提高了43.94%、33.76%、29.41%、28.61%、27.40%、26.42%、25.59%和20.40%的突变株。结论利用高通量筛选方法可以快速获得多杀菌素高产菌株。Objective High throughput screening method was employed to generate mutations in Saccharopolyspora spinosa in order to obtain stable spinosad over-production strain. Methods NTG was used to mutate S. spinosa ASAGF73 strain with a 96-weU plate and bioassay for high throughput screening. Results The positive mutants with larger proportion were obtained when the spores of the original strain were treated by 2mg/mL NTG for 50min. Eight mutants have been obtained through three rounds screening and they showed enhanced spinosad production by 43.94%, 33.76%, 29.41%, 28.61%, 27.40%, 26.42%, 25.59%, 20.40%, compared to original strain. Conclusion High throughput screening is a fast and efficient method for obtaining stable spinosad over- production strain.
