摘 要:目的探讨聚合人脐带血血红蛋白(PolyPHb)预处理对SD大鼠肝脏缺血/再灌注(I/R)损伤的保护作用。方法采用随机分组对照研究的方法,将30只成年雄性SD大鼠平均分为假手术(Sham)组、I/R组和PolyPHb预处理(PolyPHb)组;采用大鼠70%肝I/R损伤模型,缺血前I/R组和PolyPHb组动物分别静脉推注生理盐水和PolyPHb(0.3 gHb/kg)预处理,然后缺血60 min,再灌注3 h。检测术前及术后丙氨酸转氨酶(ALT)和谷草转氨酶(AST)活性,并采集术后肝脏组织标本行HE染色和TUNEL染色。结果 PolyPHb组、I/R组的肝酶释放ALT、AST分别为(350.9±80.8)U/L vs(830.5±85.7)U/L(P<0.01)和(446.1±97.10)U/L vs(1 096.2±135.6)U/L(P<0.01),肝细胞凋亡发生率分别为(15.62±8.58)%vs(35.21±9.62)%(P<0.01)。结论 PolyPHb预处理对大鼠肝脏I/R损伤具有明显的保护作用。Objective To investigate the effect of polymerized human placenta hemoglobin (PolyPHb) pretreatment on rat after hepatic ischemia/reperfusion (I/R) injury. Methods Thirty adult male SD rats were randomly divided ( n = 10 per group) to Sham group,control group (I/R) or PolyPHb pretreatment group (PolyPHb). A rat model of 70% hepatic I/R injury was used. Before ischemia, rats in the I/R group and PolyPHb group were intravenously injected with physiological saline and PolyPHb (0. 3 gHb/kg), respectively. Then the rats were subjected to 60 min hepatic ischemia and 3 h reperfusion. The releases of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) before ischemia and 'after reperfusion were detected. Liver tissues were harvested for hematoxylin-eosin (HE) staining and TUNEL staining. Results As compared to the I/R group, pretreatment with PolyPHb significantly reduced the releases of ALT, which was (350. 9 v 80.8) U/Lvs (830.5 ±85.7) U/L(P〈0. 001), and AST (446.1 ±97.1) U/Lvs (1 096.2 ±135.6) U/L(P〈 0. 001 ) ,decreased liver histopathologic changes and decreased hepatic apoptosis ( 15. 62 ± 8. 58) % vs (35.21 ±9. 62) % (P 〈0. 01 ). Conclusion Pretreatment with PolyPHb provided profound protection on rat liver against I/R injury.
