IGF-Ⅰ、GLP-Ⅰ对体外培养脂肪细胞HSL酶活性的影响

摘　　要：取体外培养单层生长良好脂肪细胞,培养介质中分别添加0、10、20、30、40、50μg/L的胰岛素样生长因子Ⅰ(IGF-Ⅰ),0、100、250、500、750、1000nmol/L的胰高血糖素样肽Ⅰ(GLP-Ⅰ),培养24h后提取总蛋白,每个处理3个重复,分别采用脂肪酶测定试剂盒检测HSL酶活性的影响。结果表明,IGF-Ⅰ、GLP-Ⅰ抑制脂肪细胞内HSL酶活性,存在剂量依赖性。A test was designed to investigate whether IGF-I,GLP-I affect the activity of HSL in adipose cells from a newborn Holstan calf. IGF-I, GLP-I were added to the media with 0,10,20,30,40,50 μg/L, 0,100,250,500,750,1 000 nmol/L,respectively. Activity of HSL in adipose cells was determined by the Kit of Lipase Activity Assay. The results showed that the activity of HSL was suppressed in adipose cells treated with IGF-I,GLP-I respectively. This suppression was dose dependent.
