摘 要:目的探讨菟丝子含药血清对SD大鼠胚胎肢芽软骨发育的影响。方法采用中药血清药理学方法制备大鼠菟丝子含药血清。取妊娠第15天SD孕鼠胚胎前肢芽,随机分为3个实验组和阴性对照组,采用肢芽悬浮培养法。实验组分别加入高、中、低剂量菟丝子含药血清(每日按照40g/kg、20g/kg、10g/kg的剂量灌胃),对照组加入正常血清。体外培养3d后收获肢芽,阿里新兰染色后采用Neubert评分系统进行评分。结果加入低剂量、中剂量、高剂量菟丝子含药血清培养的各组SD大鼠前肢芽Neubert评分的总分依次为223.49±3.26、218.89±3.69、203.21±3.62。阴性对照组Neubert总分为218.28±3.78。其中低、中剂量组的肢芽评分与对照组比较无显著差异(P>0.05)。而高剂量组肢芽评分明显低于其它各组与对照组比较有显著差异(P<0.05)。结论高剂量菟丝子含药血清对SD大鼠胚胎前肢芽软骨的发育有一定抑制作用。Objective: To observe the influence of Cuscuta chinensis Lam medicated serum on embryos limb buds cartilage devel- opment of SD rats in vitro. Methods : The differet dosages Cuscuta chinensis Lam medicated serum were prepared by serological phar- macological method. The forelimb buds were amputated from SD rat embryos on dayl5. Four test groups of amputated rat forelimb buds included the experimental groups which were cultured separately by low, middle, high dosages Cuscuta chinensis Lam medicated serum and the blank control group which was cultured by nothing Cuseuta chinensis Lam medicated serum. The limb buds were cultured in vitro for 3 days and stained with alcian blue, then evaluated by Neubert score. Results: The Neubert scores of forelimb buds which were cultured by the low and middle dosages Cuscuta chinensis Lam medicated serum were 223.49 ± 3.26 and 218. 89 ± 3.69, which had no evidently difference with the blank control group that got 218.28±3.78 (P 〉0. 05). However, the Neubert scores of forelimb buds which were cultured by the high dosages Cuscuta chinensis Lam medicated serum were 203.21 ± 3.62, which had evidently differ- ence with the blank control group ( P 〈 0.05 ). Conclusion : The high dosage Cuscuta chinensis Lain medicated serum could inhibit the development of rat embryos forelimb buds cartilage.
