摘 要:鸭坦布苏病毒(DTMUV)为新出现的病毒,主要引起鸭的产蛋量急剧下降,甚至绝产。为评价其灭活疫苗的免疫效力,本实验从产蛋下降综合征的病鸭中分离鉴定到一株DTMUV,经鸭胚增殖,测定其尿囊液中病毒含量为5×102.25ELD50/mL;采用该尿囊液按照常规方法制备灭活油乳苗。将10日龄雏鸭分成4组,每组20只,分别免疫0.2 mL,0.5 mL,0.8 mL疫苗,以及PBS对照组,并于免疫后28 d攻毒。通过间接ELISA法检测鸭血清中抗体水平,结果显示,在免疫后7 d~21 d内,其中0.5 mL免疫组抗体水平与对照组相比差异极显著(p<0.01);免疫21 d时,E玫瑰花环试验检测鸭血清中T淋巴细胞数量,结果 0.5 mL组玫瑰花环形成率为67.09±1.23%,极显著(p<0.01)于对照组(51.86±1.14%);攻毒保护试验结果显示,0.2 mL组免疫保护率为80%,0.5 mL和0.8 mL组疫苗保护率均为100%,而对照组雏鸭均表现为典型的DTMUV感染症状,其中3只死亡。实验结果表明,本研究制备的鸭坦布苏病灭活疫苗能够有效诱导抗体和细胞免疫反应,为进一步研究鸭坦布苏病疫苗提供了实验数据。Duck Tembusu virus (DTMUV) was newly emerged which causes severe egg-drop syndrome in duck. In this study, a DTMUV was isolated from sick duck with egg-drop syndrome and the inactivated oil emulsion vaccine was prepared with the virus in a titer of 5x10225 ELD^mL of duck embryo allantoic fluid by the conventional methods. Ducklings at 10 day old were divided into 4 groups (n=20) and immunized with the vaccine at the dosage of 0.2, 0.5 and 0.8 mL per duckling in vaccinated groups via intramuscular injection, with a unvaccinated control group and challenged at 28 days post immunization (PI). In addition, the antibody level was highly significant (ao〈0.01) in 0.5mL group to control group within 7 to 21 days PI, detected using the ELISA method. The E rosette forming rate in 0.5mL group (67.09~1.23%) was also highly significant (p〈0.01) to the control group (51.86±1.14%) at 28 days PI. Moreover, the protection rates of the vaccine were 80% in 0.2 mL group and 100% in both 0.5 and 0.8mL groups, respectively, whereas, all duckings in control group shown the typical clinical signs with 3 died post challenge. In conclusion, the results indicated that the inactivated oil emulsion vaccine of DTMUV was able to efficiently induce the humoral and cellular immunity against DTMUV infection.
