摘 要:目的:观察跑台运动对大鼠骨骼肌急性钝挫伤后修复过程中转化生长因子β1(Transforming growth factor-β1,TGF-β1)和碱性成纤维细胞生长因子(Basic fibroblast growth factor,bFGF)表达的影响。方法:雄性Sprague-Dawley大鼠33只,随机分为正常对照组(A组,n=3)、自然愈合组(B组,n=15)和跑台运动组(C组,n=15)。A组于建模前全部处死取材(6肢)。B组与C组用自制重物坠落打击装置造成双侧后肢腓肠肌中段闭合性急性钝挫伤,建立骨骼肌急性钝挫伤模型(60肢)。建模后B组不施加运动干预,自然愈合;C组每天进行定时定量跑台训练。B组和C组分别于伤后第3、6、9、12、21天随机选取3只(6肢)处死,在击打中心部位取材制成病理切片,行免疫组化染色,并使用Imagepro plus 6.0图像分析软件进行定量分析,计算TGF-β1和bFGF的平均光密度,观察其表达水平。结果:A组仅有微量TGF-β1和bFGF表达。B组伤后3天TGF-β1、bFGF平均光密度值开始增高,6天达到峰值,9天开始逐渐下降,两因子表达特点相似。B、C两组伤后各取材时间点两因子的表达均显著高于A组(P<0.01)。C组伤后各取材时间点TGF-β1平均光密度值显著高于B组(P<0.01);伤后3、6、9、12天bFGF平均光密度值均升高(P<0.01或P<0.05),3天时达峰值,21天时无显著性差异(P>0.05)。结论:骨骼肌急性钝挫伤后,早期施加运动干预促进TGF-β1和bFGF表达,提示骨骼肌愈合过程中可能发生瘢痕过度增生而影响愈合质量。Objective To investigate the effect of treadmill exercise on the expression of transforming growth factor-βl (TGF-βl)and basic fibroblast growth factor (bFGF) in skeletal muscles after acute contusion. Methods Thirty-three adult male Sprague-Dawley (SD) rats were randomly divided into 3 groups : normal control group (group A, n =3), spontaneous closure group (group B, n =15), treadmill exercise group (group C, n=lS). The rats in group A were sacrificed and bilateral gastrocnemius muscle was isolated. Contusion of bilateral gastrocnemius was simulated in groups B and group C. The rats in group C took part in treadmill exercise every day after injury,while the rats in group B administered no any intervention. The rats in group B and group C were sacrificed 3,6,9,12 and 21 days after injury, respectively,and their bilateral gastrocnemius were isolated. Immunohistochemical analysis was used to measure the expression of TGF-[31 and bFGF at different time points. Imagepro plus 6.0 image analysis software was used for quantitative analyzing. Results The expressions of TGF-[31 and bFGF in groups B and C increased significantly when compared with those in group A (P 〈 0.01 ), The expression of TGF- [3l significantly increased in group C when compared with that in group B (P 〈 0.01 ) 3,6,9,12 and 21 days after contusion. The expression of bFGF increased significantly in group C when compared with that in group B (P 〈 0.05 at 6th,9th and 12th day after contusion,P 〈 0.01 at 3th day after contusion). Conclusion Increased expression of TGF-151 and bFGF induced by early exercise after muscle contusion could promote the development of fibrosis and scar formation and eventually hinder the functional recovery of injured skeletal muscle.
