摘 要:采用生化分析方法,研究了土荆芥根系分泌物对大豆根尖细胞的氧化损伤.结果表明,在土荆芥组培根分泌物处理下,大豆根尖细胞O2.-和H2O2水平升高,丙二醛(MDA)含量增大,其中,100和1 000μg/mL处理组活性氧自由基(ROS)水平升高达到显著程度;随着处理时间延长,根尖细胞的超氧化物歧化酶(SOD)活性和过氧化物酶(POD)活性在1和10μg/mL处理组持续升高,而在100和1 000μg/mL处理组则呈先升高后降低的趋势.过氧化氢酶(CAT)活性变化规律表现出持续升高的态势,在处理24 h后达到最大.电泳分析结果显示处理未造成大豆根尖细胞产生DNA片段.土荆芥组培根分泌物未导致大豆根尖细胞凋亡.This work investigated the oxidative damage of cultured Chenopodium ambrosioides root exudates on root tip ceils of soy- bean through biochemical analysis. The results showed that tissue cultured C. ambrosioides root exudates could stimtrlate the oxygen production of root tip cells of soybean and increase the concentration of MDA. There was a significant increase of ROS level in the treat- ment groups of 100 and 1 000 μg/mL. Along with the treatment time, the SOD and POD activities of root tip cells increased continu- ously in lower concentration ( 1 and 10 μg/mL). Whereas in high concentration treatment group ( 100 and 1 000 μg/mL) , the activi- ties increased at first and dropped latter. The activity of CAT was increasing stably and reached the maximum level at 24 h after treat- ment. There was no DNA break of root tip cells of soybean in the treatment according to the analysis of electrophoresis. Cultured C. ambrosioides root exudates did not induce apoptosis of soybean root tip cell.
