摘  要:对核糖体DNA第2内转录间隔区(rDNA-ITS2)序列作为中国按蚊属按蚊亚属蚊种鉴别的分子特征进行评价。笔者整理和分析了GenBank数据库中注册的所有中国记录的按蚊亚属蚊种的rDNA—ITS2序列,并应用MEGA软件计算其在种内和种间的差异性(P距离)。截止2012年10月,GenBank中已注册ITS2序列共584条,隶属按蚊亚属蚊20种(包括同物异名)、待定种5种和杂交种1个。计算结果显示,ITS2序列在种内的变异性小于1%,种间的差异性范围为1.2%(凉山按蚊与昆明按蚊)-69.5%(待定种YM.2003a与须喙按蚊)。另外,发现少数注册的序列在种内差异性较大,在比对序列时需谨慎。结果提示rDNA-ITS2序列在本研究按蚊中具有良好的种内保守性和种间解析度。To evaluate the ribosomal DNA second internal transcribed spacer (rDNA-ITS2) sequences as a identification identity of Chinese genus Anopheles subgenus Anopheles mosquito species. The rDNA-ITS2 sequences of Chinese subgenus Anopheles mosquito species registered in the GenBank database were downloaded and organized. The intraspecific variation and interspecific difference (p-distance) were estimated by MEGA software. By October 2012, there were 584 of ITS2 sequences registered on GenBank, which belong to 20 species (including synonymy) , 5 unnamed species and 1 hybrid. The value of intraspecific p-distance was less than 1%, and the range of interspecific p-distance as from 1.2% (An. liangshaneneis with An. kunmingensis) to 69.5% (An. sp YM-2003a with An. barbirostris). In addition, intraspecific variation of a few sequences was considerable, which need to be careful in the blast searching. The result showed that the rDNA-ITS2 sequence has a good conservation and interspecific resolution in this anopheles mosquito.
