摘  要:目的探讨孕鼠孕期低剂量十溴联苯醚(BDE-209)暴露联合窒息对新生仔鼠海马组织丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性的影响。方法 12周龄SD雌性大鼠自妊娠0 d起随机分成A组:空白对照,B、C组:0.6、6 mg/kg BDE-209暴露组,D组:孕期窒息15 min,E、F组:0.6、6 mg/kg BDE-209暴露+孕期窒息15 min。B、C、E、F组雌鼠从孕0天开始胃灌BDE-209,A、D组胃灌等量的花生油直至孕第21天(即孕22 d),A、B、C组孕鼠行剖宫产术直接取出仔鼠,D、E、F组孕鼠行剖宫产建立孕期窒息模型后再取出仔鼠。各组随机选取10只雄性仔鼠,在出生24 h内检测海马MDA含量及SOD活性。结果各实验组仔鼠MDA含量随BDE-209暴露剂量增加而增高,SOD活性随BDE-209暴露剂量增加而降低,在BDE-209暴露剂量相同的情况下,孕期窒息能够增加MDA含量,降低SOD活性,组间两两比较,差异有统计学意义(P=0.000)。孕期低剂量BDE-209暴露联合窒息对SOD活性的影响存在交互作用(P=0.002)。结论孕期低剂量的BDE-209暴露联合窒息可以使仔鼠海马SOD活性下降,MDA含量升高,出现氧化应激状态,导致神经系统的氧化损伤。孕期低剂量BDE-209暴露与孕期窒息之间存在一定的交互作用。Objective To study the effects of pregnant rats in perinatal period exposed to low dose BDE-209 combined with asphyxia on the content of MDA and activity of SOD in neonatal rats hippocampus. Methods Twelve week-old female SD rats were randomly divided into 6 groups from the first day of their gestation: group A, the blank control group; group B,0.6 mg/kg BDE-209; group C, 6 mg/kg group E,0.6 mg/kg BDE-209 + asphyxia neonatorum for 15 BDE-209; group D, asphyxia neonatorum for 15 minutes; minutes;group F, 6 mg/kg BDE-209 + asphyxia neonatorum for 15 minutes. The female rats of group B, C, E and F were given a garage of BDE-209 from the first day of gestation to the 21st days. Group A and D were given a gavage of peanut oil of the same volume with BDE-209 from the first day of gestation to the 21st days. Then rats in group A, B and C underwent cesarean section directly to obtain offspring rats, the ones in group D, E and F underwent cesarean section after established asphyxia neonatorum model. We selected 10 newborn male rats of each group randomly, and detected the content of MDA and activity of SOD in 24 hours after birth. Results In these newborn male rats, the content of MDA increased with the dose of BDE-209 increasing, while the activity of SOD reduced with the dose of BDE-209 increasing. Asphyxia neonatorum could increase the content of MDA and reduce the activity of SOD with the same dose BDE-209 exposure. The difference between these groups was statistically significant (P=0.000). There were interaction between exposing on low dose of BDE-209 and asphyxia neonatorum on the activity of SOD (P=0.002). Conclusions Perinatal exposure on low dose of BDE-209 combined with asphyxia can decrease the activity of SOD, increase the content of MDA in rat hippocampus and cause the oxidative stress and oxidative damage. There are interactions between exposing on low dose of BDE-209 and asphyxia neonatorum.
