摘  要:目的通过研究旋毛虫感染鼠在过敏性哮喘发病时血清及支气管肺泡灌洗液中IL-4和IL-5的水平变化,探讨旋毛虫感染对过敏性哮喘的作用。方法取雌性、8周龄BALB/c小鼠20只,随机分为4组,A组为空白对照组,B组为单纯过敏性哮喘组,C组为感染旋毛虫后哮喘组,D组为单纯感染旋毛虫组。C、D组小鼠经灌胃法感染旋毛虫囊包幼虫200~300条;4周后,以卵清白蛋白(OVA)分别对B、C组小鼠进行致敏激发,建立过敏性哮喘模型;8周后,取小鼠血清、肺泡灌洗液,用ELISA法检测IL-4和IL-5水平。结果血清中A、B、C、D组小鼠IL-4水平分别为(136.4948±31.1670)、(209.8004±21.5652)、(162.7892±17.6829)、(182.7092±12.0841)pg/L;IL-5水平分别为(7.3101±1.8600)、(15.9997±2.28208)、(9.2411±1.7280)、(11.4135±2.10776)ng/L;与B组比较,C组小鼠IL-4、IL-5水平均降低,差异有统计学意义(P<0.05)。肺泡灌洗液中A、B、C、D组小鼠IL-4水平分别为(169.1636±9.9997)、(232.9076±17.9501)、(172.3508±19.1895)、(193.0676±16.5708)pg/L;IL-5水平分别为(12.8618±1.7799)、(23.8460±5.507)、(15.7560±4.5419)、(20.3440±3.9713)ng/L;与B组比较,C组小鼠IL-4、IL-5水平降低,差异有统计学意义(P<0.05)。结论旋毛虫感染对过敏性哮喘小鼠IL-4、IL-5的表达有抑制作用。Objective To investigate the effect of Trichinella spiralis infection on allergic asthma, through the study of the change of IL-4 and IL-5 levels in serum and bronchoalveolar lavage fluid (BALF) when the mice were suffering from allergic asthma. Methods Twenty BALB/c female mice (8 weeks old) were randomly divided into four groups of five mice each, including A as control group, B as allergic asthma group, C as Trichinella spira!is infection followed by induced asthma group, and D as Trichinella spiralis infection group. The animals in group C and D were infected with 200~300 Trichinella spiralis muscle larvae by lavage. Four weeks later, Ovalbumin(OVA) was used to induce the allergic asthma for mice in group B and C, in order to establish experimental animal model. After eight weeks, all mice were killed and IL-4 and IL-5 levels of serum and BALF were measured by ELISA. Results IL-4 serum levels of group A, B, C and D were (136.4948±31.1670), (209.8004±21.5652), (162.7892±17.6829), and (182.7092±12.0841)pg/L, respectively. IL-5 serum levels of group A,B,C and D were (7.3101±1.8600), (15.9997±2.2820), (9.2411±1.7280), and (11.4135±2.10776)ng/L,respectively. Compared with group B, IL-4 and IL-5 levels in group C decreased (P〈 0.05). BALF IL-4 levels in group A,B,C and D were (169.1636±9.9997), (232.9076±17.9501), (172.3508± 19.1895), and (193.0676±16.5708)pg/L, respectively. BALF IL-5 levels in group A, B, C and D were (12.8618± 1.7799), (23.8460±5.507), (15.7560±4.5419) and (20.3440±3.9713 ) ng/L, respectively. Compared with group B, IL- 4 and IL-5 levels in group C decreased(P〈0.05). Conclusion Trichinella spiralis infection can inhibit the expression of IL-4 and IL-5 of mice with allergic asthma.
