摘 要:[目的]观测脾阴虚大鼠模型体内氧自由基损伤程度和端粒长度。[方法]使用随机平行对照方法,将40只健康SD雌性大鼠按随机数字表方法随机分为两组(20只/组)。采用"饮食不节+疲劳过度"复合方法复制脾气虚大鼠模型,再以药物损伤法,使用温热伤阴药灌胃10d复制脾阴虚模型。观测大鼠血清丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)和脑皮质端粒长度含量。[结果]SOD活性,GSH-Px活性,T-AOC和MDA含量脾阴虚模型组与正常组相比较均有显著差异(P<0.05)。脑皮质中端粒长度模型组明显缩短(P<0.05)。[结论]脾阴虚模型大鼠存在明显自由基氧化损伤和端粒长度变短,进一步验证中医脾虚衰老学说,脾阴虚处于脾虚病程哪种阶段尚需深入研究。[ Objective ] This research was to explore the changing rule in the spleen yin deficiency rats' are attacked by free radicals and the change of the telomeric length. Methods : Used method of Randomized controlled, 40 healthy female SD rats were randomly divided into normal control group and model group (20/ group). "Fatty diet + over fatigue" method to established the model for 14 days, then gived the rats with shangyin drug for 10 days and measured the SOD, GSH - Px, T - AOC, MDA content in brain cortex and serum, measured the telomerie length in brain cortex. Results : In serum and cerebral cortex, the content of SOD, GSH - Px ,T - AOC and MDA in the model group showed a significant difference ( P 〈 0.05 ). The telomeric length in brain cortex of the model group increased shorten significantly ( P 〈 0.05 ). Conclusion : There is a clearly damage of oxidative free radical and telomeric length shorten significantly in spleen yin deficiency model, and proving the TCM theories of aging further, but the spleen yin deficiency state in what stage of the spleen desease is needed to understand in -depth studies.
