摘  要:目的探讨天然药物Serrin B和Nodosin对人早幼粒白血病细胞株HL60、人胃腺癌细胞株SGC7901、人结肠癌细胞株LOVO、人胶质瘤细胞株U87、人肺腺癌细胞株AGZY的毒性作用。方法将对数期生长的HL60、SGC7901、LOVO、U87、AGZY细胞株接种于96孔板中,细胞贴壁后,分别加入Serrin B或Nodosin,使药物的终浓度分别为0.00、1.25、2.50、5.00、10.00、20.00μmol.L-1,24 h后用四甲基偶氮唑盐法检测细胞株的死亡率。结果 Serrin B对HL60、SGC7901、LOVO、U87和AGZY细胞株均有较强的毒性作用(P<0.01),且其毒性作用呈现剂量-效应关系。Nodosin对HL60、SGC7901、LOVO、U87细胞株具有较强的毒性作用(P<0.05,P<0.01),而对AGZY细胞株无明显毒性作用。结论 Serrin B和Nodosin是中药溪黄草抗癌作用的有效成分,为人类多种肿瘤治疗的新药研究开发提供新的思路。Objective To study the toxic effects of Serrin B and Nodosin on human promyelocytic leukemia cell line HL60, human gastric adenocarcinoma cell line SGC7901, human colon carcinoma cell line LOVO, human glioma cell line U87 and human lung adenocarcinoma cell line AGZY in vitro. Methods The HL60, SGC7901, LOVO, U87 and AGZY cell lines in logarithmic phase were seeded in 96-well plates. When the cells adhere to the plates, different concentrations of Serrin B or Nodosin were added to the wells. The final concentrations were 0. 00,1. 25 ,2. 50,5. 00,10. 00,20. 00 μmol . L ^- 1 respectively. 24 hours later, four methyl thiazolyl tetrazobium methods were used to determine the death rates of the ceils. Results There were obvious toxic effects for Serrin B on HL60, SGC7901, LOVO, U87 and AGZY cell lines in vitro(P 〈0.01 ). The dose-effect relationship was obvious. There were significant toxic effects for Nodosin on HL60, SGC7901, LOVO, U87 cell lines ( P 〈 0.05,P 〈 0.01 ). There was no toxic effect for Nodosin on AGZY cell lines. Conclusion Serrin B and Nodosin are the active anticancer components of Robdosia serra (Maxim) Hara, which offers more clues for the development of new antican- eer drugs.
