摘 要:目的:建立HPLC法测定大青根中腺苷和丁香树脂酚葡萄糖苷含量的方法。方法:采用Diamnosil ODS-C_(18)色谱柱(200 mm×4.6 mm,5μm),流动相为乙腈-水梯度洗脱,检测波长为254 nm,流速为1.0 ml·min^(-1),柱温为室温,进样量为10μl。结果:腺苷和丁香树脂酚葡萄糖苷线性范围分别为5~100μg·ml^(-1)(r=0.999 6),5~100μg·ml^(-1)(r=0.999 3);平均回收率分别为98.2%(RSD=1.08%,n=6)和98.4%(RSD=1.18%和n=6);样品平均含量分别为0.081,0.014 mg·g^(-1)。结论:本方法可用于大青根药材的质量控制。Objective:To establish an HPLC method for the simultaneous determination of adenosine and syringaresinol-4-O-β-Dglucopyranoside in the root of Clerodendron cyrtophyllum Turcz.Method:A Diamnosil ODS-C_(18) column(200 mm×4.6 mm,5μm) was applied using a mixture of acetonitrile-water as the mobile phase with gradient elution.The flow rate was 1.0 ml·min(-1) and the column temperature was maintained at the room temperature.The detection wavelength was 254 nm.The sample volume was 10μl.Result: The linear range of adenosine and syringaresinol-4-O-β-D-glucopyranoside was 5-100μg·ml(-1)(r =0.999 6) and 5-100μg·ml(-1)(r=0.9993),respectively.The average recovery was 98.2%with RSD of 1.08%(n =6) for adenosine and 98.4%with RSD of 1.18%(n =6) for yringaresinol-4-O-p-D-glucopyranoside.Conclusion:The method is accurate and reproducible,which can be used in the quality control of the root of Clerodendron cyrtophyllum Turcz.
