摘　　要：目的用纯化人sCR1蛋白作抗原免疫家兔,制备sCR1多克隆抗体.方法培养E. coli BL21克隆菌,采用IPTG诱导表达sCR1蛋白,在变性条件下纯化、复性,将其作为抗原免疫家兔,制备兔抗人sCR1多抗,并进行SDS电泳鉴定分析.结果在E. coli BL21中高效表达人sCR1蛋白,有较好的免疫原性,获得了高效价的兔抗人sCR1抗体,可特异地识别人sCR1蛋白.结论纯化的sCR1蛋白免疫兔子后,制出的兔抗人sCR1多克隆抗体效价较高,为免疫学检测方法的建立奠定了基础.Objective The human sCR1 protein was used as the immunogen after purified, The rabbit antibody against human sCR1 was successfully prepared. Method The recombinant human sCR1 fusion protein was expressed in E. coli BL21（DE3）, with IPTG induction, after purified and refolding, then used as the immunogen to immunize the rabbit. Properties of antiserum against human sCR1 were identified by ELISA, after purification by immunoaffinity chromatography and verified by SDS-PAGE. Result The polyclonal antibody with high titer and high specificity was obtained, which could recognize human sCR1 protein specifically. Conclusion The human sCR1 fusion protein was used as the immunogen after purified and refolding, which has better antigenicity and immunogenicity. This research lays a foundation for further research into the detection and functional study of human sCR1.
