摘  要:目的:观察胞壁酰二肽对人主动脉内皮细胞数种炎症因子表达的影响。方法:用胞壁酰二肽(MDP)按要求对人主动脉内皮细胞(HAECs)进行处理,运用实时定量RT-PCR以及RT-PCR测定IL-1β,IL-8,MCP-1,IFN-α与IFN-β基因的表达。用Western blot与RT-PCR方法研究MDP影响IFN-α,IFN-β表达的机制。结果:经MDP处理后,HAECs细胞中炎症因子IFN-α,IFN-β,IL-1β,IL-8和MCP-1的表达上调。结论:MDP可以通过上调HAECs细胞炎症因子的表达。Objective: To observe effects of muramyl dipeptide on the expression of inflammatory cytokine in human aortic endothelial cells. Methods. Human aortic endothelial ceils (HAECs) were trea- ted with muramyl dipeptide (MDP) according to the requirements, the gene expression of IL-1β, IL-8, MCP-1 IFN-α and IFN-β were measured by RT-PCR and real time RT-PCR method. RT-PCR and Western blot were used to explore the mechanism of the up-regulated of IFN-β and IFN-β. Results: After muramyl dipeptide (MDP) treatment, the expression of IL-1β, IL-8, MCP-1 IFN-β and IFN-β were up-regulated. Conclusion: MDP can injure endothelial function by up-regulate the expression of inflammatory eytokine.
