摘 要:目的研究壳聚糖和三聚磷酸钠(TPP)包载EPO的纳米粒对受伤性脑损伤(TBI)大鼠的作用。方法用离子交联法,制备壳聚糖(chitosan,CS)-三聚磷酸钠(TPP)/EPO纳米粒,对纳米粒进行表征。TBI大鼠分成4组(每组8只),用PBS、1 mg.mL-1壳聚糖(CS)-TPP 300μL、200 U.mL-1EPO 300μL和CS-TPP/EPO 300μL分别皮下注射,用水迷宫法评价其对脑损伤的作用。结果壳聚糖-三聚磷酸钠/EPO纳米粒的粒径(348±8)nm,EPO的包封率为75.16%,在pH 7.4的释放曲线成典型的双相释放。载EPO纳米粒对TBI大鼠作用明显优于对照组。结论壳聚糖-三聚磷酸钠/EPO纳米粒能显著改善TBI的损伤。Objective To study the protective effects of loaded erythropoietin (EPO) with chitosan/tripolyphosphate nanoparticles in traumatic brain injury (TBI) model. Methods Chitosan ( CS ) - tripolyphosphate (TPP)/EPO nanoparticles were subcutaneous injection in prepared using ionic gelation, and were characterized. TBI model were divided into four groups(all n =8). PBS, 1 mg · mL^-1 CS -TPP 300 μL, 200 U · mL-1 EPO 300 μL and CS -TPP/EPO 300 μL nanoparticles were iv 333I rats and the efficacy was evaluated using Morris water maze test. Re- suits The size of CS - TPP/EPO nanoparticles was (348 ± 8) nm, which was further verified by scanning electron microscope (SEM). EPO loading efficiency of CS/TPP nanoparticles was 75. 16%, and the EPO release of nanoparticles was bi - phase release at pH 7.4.
