金匮肾气丸对肾阳虚变应性鼻炎缓解期小鼠Th1/Th2细胞因子的影响

摘　　要：目的观察金匮肾气丸对肾阳虚变应性鼻炎(AR)缓解期小鼠Th1/Th2细胞因子的影响,探讨中医在AR缓解期"治未病"的可能机理。方法建立肾阳虚AR小鼠模型,实验分肾阳虚AR组、金匮肾气丸组和正常对照组。采用酶联免疫吸附法测定小鼠血清中Th1/Th2细胞因子和组胺,并观察小鼠鼻黏膜肥大细胞、嗜酸性粒细胞浸润情况。结果金匮肾气丸能改善肾阳虚AR模型小鼠鼻黏膜的变态反应症状;降低肾阳虚AR小鼠血清IL-4水平,升高IFN-γ、IL-2水平,降低组胺含量(P<0.01)。结论金匮肾气丸对肾阳虚AR小鼠的治疗作用可能与其下调小鼠血清IL-4,上调IFN-γ、IL-2水平,纠正失衡的Th1/Th2有关。Objective To observe the effects on Jinkui Shenqi pills, which is for tonifying-kidney yang, on Th1/Th2 cytokines in mice with kidney-yang deficiency allergic rhinitis（AR） in remission stage, and to explore the possible mechanism of the theory of preventive treatment of disease for AR in remission stage. Methods We set up the kidney-yang deficiency AR mice model with ovalbumin and adenine. The mice were divided into model group, Jinkui Shenqi Pills group and normal control group. Enzyme linked immunosorbent assay was adopted for the observation of cytokines and histamine in serum, and we also observed the nasal symptoms, and the infiltration of mast cells and eosnophils in nasal mucosa. Results Jinkui Shenqi Pills relieved allergic reaction of mice nasal mucosa, decreased serum IL-4 and histamine,increased IFN-γ and IL-2 cytokines in AR mice（P〈0.01）. Conclusion The therapeutic mechanism of Jinkui Shenqi Pills may be related with down-regulation of serum IL-4, up-regulation of IFN-γ and IL-2, and restoring Th1/Th2 cytokines network imbalance in kidney-yang deficiency allergic rhinitis mice.
