摘  要:以芍药去皮种子和胚为外植体,研究了不同培养基对胚萌发率和生长状况的影响。结果表明:在诱导种子萌发过程中,3种基本培养基中以MS的效果最好,且在MS+6-BA 2.0mg/L+NAA 0.2mg/L培养基上萌发率最高,达71.4%;离体胚培养中,单独添加2.0mg/L 6-BA时,胚生长受到抑制;MS+6-BA 1.0mg/L+NAA 0.4mg/L能促进愈伤组织形成;适宜浓度的GA3能够打破上胚轴休眠,MS+6-BA 1.0mg/L+GA30.5mg/L为胚诱导真叶抽生的最佳培养基。Paeonia lactiflora Pall. peeled seeds and embryos were used as explants to study the effects of different mediums on the germination and growth of Paeonia lactiflora Pall.. The results showed that MS medium was the best for inducing the germination of seeds among 3 medias and seeds had the highest germination rate (71.4%) on MS+6-BA 2. 0 mg/L+NAA 0. 2 mg/L. In vitro embryo culture,adding 6-BA solely at 2. 0 mg/L inhibited the growth of embryo; MS+6-BA 1.0 mg/L+NAA 0. 4 mg/L promoted the formation of callus;GA3 at suitable concentration could break the dormancy of epicotyl and the optional medium for inducing leaves to throw out from embryo was MS+6-BA 1.0 rag/L+ GA3 0. 5 m4g/L.
