摘 要:目的探讨局部应用油酸对糖尿病创面炎症反应和组织愈合的影响。方法 84只C57BL/6小鼠经链脲霉素诱导建立糖尿病模型,建模后1个月于小鼠背部制作直径1 cm的圆形全层皮肤缺损创面,根据创面处理方式的不同将小鼠分为油酸组(30mmol/L油酸溶液)和对照组(使用等体积溶剂代替油酸溶液),每组各42只。以数码照相获取图像观察创面愈合情况,计算创面愈合率;采集创面标本经免疫组织化学染色结合计算机图像软件计数创面内中性粒细胞(PMN)和巨噬细胞(Mφ);利用ELISA法测定创面组织中肿瘤坏死因子α(TNF-α)和白细胞介素-6(IL-6)的表达。结果从伤后5 d起,油酸组创面愈合率均显著高于对照组(P<0.01)。油酸组和对照组创面愈合时间分别为(17.2±2.6)d和(19.5±1.9)d(P<0.05)。油酸组伤后12、24 h的创面PMN计数和伤后3、5 d的创面Mφ计数均显著高于对照组(P<0.01),伤后10、15 d时创面PMN计数和Mφ计数则明显低于对照组(P<0.05或P<0.01)。油酸组伤后3 d时创面TNF-α表达及伤后12、24 h和3 d时创面IL-6表达均明显高于对照组(P<0.05或P<0.01),伤后10 d时创面TNF-α和IL-6表达均显著低于对照组(P<0.05或P<0.01)。结论油酸能够在创伤早期加速糖尿病创面的早期炎症反应进程,在后期避免炎症反应的持续,从而促进糖尿病小鼠创面的愈合。Objective To investigate the effects of topical application of oleic acid on inflammatory response and wound healing in diabetic mice. Methods The diabetic models were established by peritoneal injection of streptozocin in 84 C57BL/6 mice. One month after model establishment, full-thickness skin wounds with diameter of 1 cm were prepared in the dorsal region of mice. All the mice were divided into oleic acid group (30 mmol/L oleic acid solution) and control group (same amount of solvent instead of oleic acid solution), with 42 mice in each group. The wound sites were digitally photographed to calculate the percentage of wound closure using computer image analysis software. The wound specimens were also obtained for detecting the infiltration of polymorphonuclear neutrophils (PMN) and macrophages (Mq0) using immunohistological technique. The expression of tumor necrosis factor-alpha (TNF-ct) and interleukin-6 (IL-6) in wound tissues was determined by ELISA. Results From the fifth day after surgery, the percentages of wound closure in oleic acid group were significantly higher than those in control group (P 〈 0.01). The wound closure time in olcic acid group and control group was (17.2 ±2.6) d and (19.5± 1.9) d respectively (P 〈0.05). The levels of PMN 12 and 24 h after surgery and those of Mq~ 3 and 5 d after surgery in oleic acid group were significantly higher than those in control group (P 〈0.01), while the levels of PMN and Mq0 10 and 15 d after surgery in oleic acid group were significantly lower than those in control group (P 〈 0.05 or P 〈 0.01). The expression of TNF-ct in wound tissues 3 d after surgery and the expression of IL-6 in wound tissues 12 h, 24 h and 3 d after surgery in oleic acid group was significantly higher than that in control group (P 〈 0.05 or P 〈 0.01), while the expression of TNF-ct and IL-6 in wound tissues 10 d after surgery was significantly lower than that in control group (P 〈 O. 05 or P 〈 O. 01).
