半夏等6种中药超临界CO_2萃取物对青霉素诱发大鼠痫性放电和海马区相关递质含量的比较研究

摘  要:目的:比较半夏等6种中草药的超临界CO2萃取物对大鼠皮层定位注射青霉素诱发惊厥模型的对抗作用,并探讨其作用机制。方法:建立大鼠皮层定位注射青霉素惊厥模型,比较灌胃各药对惊厥行为和脑电图的影响;采用HPLC法比较各药对海马区Glu、Asp、Gly和GABA的含量影响。结果:(1)半Ⅱ、柴Ⅰ、天Ⅱ、钩Ⅱ、石Ⅰ、石Ⅱ均可不同程度地延长青霉素诱发痫性的发作潜伏期,减轻发作程度,延长痫性放电的潜伏期,减少痫波发放频率,减小放电最高波幅;(2)钩Ⅱ和半Ⅱ能显著降低Glu的含量(P<0.05);柴Ⅰ、石Ⅰ、天Ⅱ和半Ⅱ能显著升高GABA的含量(P<0.01或P<0.05)。结论:(1)半Ⅱ、柴Ⅰ、天Ⅱ、钩Ⅱ、石Ⅰ、石Ⅱ可抑制大鼠皮层定位注射青霉素诱发的癫痫发作和痫性放电,产生抗癫痫作用,其中半Ⅱ是对抗青霉素点燃惊厥发作最有效的药物,其作用性质与托吡酯相似,且对该模型的对抗作用优于托吡酯;(2)柴Ⅰ、石Ⅰ和天Ⅱ的抗惊厥作用机制主要通过显著增加大鼠海马GABA的含量、提高GABA功能实现,钩Ⅱ通过降低Glu的含量、限制Glu兴奋系统功能实现,半Ⅱ既能增加GABA的含量又能降低Glu的含量,其抗痫作用可能与加强GABA抑制功能和/或限制Glu兴奋功能有关。Objective:To compare the anticonvulsion effect of supercritical-CO2 fluid extractions(SFE-CO2 ) of Pinellia Tuber et al, on the mode of seizure rats induced by penicillin and explore its mechanism of anticonvulsant action. Methods : Using the model of seizure rats induced by penicillin localized injected in cortex investigated the effect of extractions on the convulsant behaviors and hippocampus EEG. The neurotransmitters (Glu, Asp, Gly and GABA)of extractions in hippocampus were determined by high performance liquid chromatography (HPLC). Results: ( 1 ) PT Ⅱ , RP Ⅱ , UN Ⅱ , RAT Ⅰ , RAT Ⅱ and RB Ⅰ can prolong the latent period of epileptiform discharge, reduce the frequency of epileptiform discharge, and decrease the highest wave of hippocampus EEG. (2) UN H and PT Ⅱ can significantly decrease the level of GIu(P 〈 0. 05) ; RB Ⅰ , RAT Ⅰ , RP Ⅱ and PT H can obviously increase the level of GABA ( P 〈 0. 01 or P 〈 0. 05 ). Conclusions : ( 1 ) PT Ⅱ , RP Ⅱ , UN Ⅱ , RAT Ⅰ and RB Ⅰ can antagonize penicillin-kindling seizure in rats. PT Ⅱ was the most effective extraction to antagonize the penicillin-induced seizure. Anticonvulsant properties of PT Ⅱ resembled Topamax, and the effect of it was better than Topamax. (2) The anticonvulsant mechanism of RB Ⅰ , RAT Ⅰ and RP Ⅱ might have been due to increase the level of GABA, however, UN Ⅱ related to decrease Glu level. The anticonvulsant mechanism of PT Ⅱ might had relation with these two aspects.
