五子衍宗方对实验性隐睾小鼠复位固定术后生精功能恢复的影响

摘　　要：目的:研究五子衍宗方对实验性隐睾小鼠复位固定术后睾丸生精功能恢复的促进作用。方法:取Balb/c小鼠随机分为假手术组、模型组、五子衍宗方100、200 mg·kg-1组,除假手术组外,其余4组分别行睾丸腹腔固定术制隐睾模型,假手术组将睾丸提至腹腔后即回复阴囊而不行固定术;术后常规饲养14 d后,对模型组及五子衍宗方组再行隐睾复位固定术,术后假手术组和模型组每天灌服纯净水0.01mL·g-1,五子衍宗方100、200 mg·kg-1剂量组分别灌服相应剂量五子衍宗方,各组小鼠连续给药14 d,于末次给药24 h后检测各组小鼠的睾丸重量、精子密度、活率,流式细胞术分析睾丸各级生精细胞周期,光、电镜观察睾丸组织学变化。结果:五子衍宗方能够提高模型小鼠睾丸重量,增加精子密度、精子活率,恢复睾丸内各级生精细胞百分比,从而改善隐睾小鼠复位固定术后睾丸组织的病理性变化。结论:五子衍宗方能够促进隐睾小鼠睾丸复位固定术后的睾丸生精功能恢复。Objective： To investigate the recovery effects of Wuzi Yanzong decoction on orchidopexied experimental cryptorchidism mice. Method： Balb/c mice were induced into cryptorchidism and divied into four groups as sham, model, low dose of Wuzi Yanzong decoction （ 100mg. kg- 1 ）, high dose of Wuzi Yanzong decoction （200mg. kg - 1 ）. Besides sham group, the rest four groups were operated to induce experimental bilateral cryptorchidism via an abdominal incision and suturing of the left and right testis to the lateral abdominal wall. Sham-operated mice were anesthetized and underwent an abdominal incision, then sutured the wound. After maintained the animals for 14 days, the model and Wuzi Yanzong decoction mice were underwent orchidopexy. The mice in low or high dose of Wuzi Yanzong decoction group were given corresponding dose once a day for 14 days starting from the orchidopexy operation. Sham and model mice were given pure water with the same volume through intragastric administration. At the end of the study, testis tissues were collected for detection of mice testes weight, sperm density,live rate, histological structure changes of testis, analysis the sperm cell cycle in testis tussues through flow cytometry. Result： The Wuzi Yanzong decoction treated mice significantly increased the mice testis weight, sperm density, sperm live rate and recover the percentage of spermatogenic cell so improved the histologic appearance. Conclusion： Wuzi Yanzong decoction is a potential beneficial agent to promote the testicular recover from dyzoospermia after orchidopexy in cryptorchid mice.
