摘 要:目的观察大鼠脑损伤后白介素-6(IL-6)及白介素-8(IL-8)的变化及地塞米松对其影响。方法将42只Sprague-Dawley(SD)大鼠按完全随机分组的方法分为:假手术组(n=6)、损伤后6h(n=6)、损伤后24h(n=6)、损伤后48h(n=6)、地塞米松干预后6h(n=6)、地塞米松干预后24h(n=6)、地塞米松干预后48h(n=6);建立大鼠液压颅脑损伤模型,假手术组不予处理,损伤组给予液压打击处理,干预组予打击后立即腹腔注射地塞米松,用酶联免疫吸附测定法测IL-6、IL-8的表达。结果损伤后6h组与假手术组比较,IL-6、IL-8表达增加,差异有统计学意义(t分别=0.01、0.02,P均<0.05)。损伤6h、24h、48h后IL-6、IL-8表达呈上升趋势,差异均有统计学意义(F分别=3.78、3.82,P均<0.05),进一步检验显示,损伤后48h和24h的IL-6、IL-8值高于损伤后6h,损伤后48h高于损伤后24h,差异均有统计学意义(t分别=0.02、0.02、0.03、0.01、0.02、0.01,P均<0.05)。干预组6h、24h、48h与损伤后比较,同时段干预组IL-6、IL-8表达均降低,差异均有统计学意义(t分别=0.04、0.01、0.02、0.02、0.01、0.01,P均<0.05)。结论地塞米松可通过抑制IL-6及IL-8的表达,达到脑保护的作用。Objective To observe the change of levels of IL-6 and IL-8 in rat brain tissue after injury and the effects after injecting dexamethasone. Methods A total of 42 Sprague-Dawley rats were randomly divided into seven groups: sham operation group, 6 hours after trauma group, 24 hours after trauma group, 48 hours after trauma group, 6 hours after Dexamethasone intervened group, 24 hours after dexamethasone intervened group and 48 hours after dexamethasone intervened group with six rats in each. Fluid percussion brain injury model of rats were established. Sham operation group was treated with nothing. Trauma groups were treated with hydraulic combat. Dexamethasone were injected immediately after trauma in dexamethasone intervented groups. The levels of IL-6 and IL-8 in brain tissue were detected by ELISA. Results The levels of IL-6 and IL-8 were significantly higher in 6 hours after trauma group than that of the sham operation group (t=0.01,0.02,P〈0.05). The levels of IL-6 and IL-8 were significantly rising at 6 hours,24 hours and 48 hours after trauma (F=3.78,3.82, P〈0.05). The levels of IL-6 and IL-8 were significantly higher at 48 hours and 24 hours than that at 6 hours after trauma. And the levels of IL-6 and IL-8 were significantly higher at 48 hours than 24 hours after trauma (t=0.02,0.02,0.03,0.01,0.02,0.01,P〈 0.05 ). Compared with trauma group at 6 hours, 24 hours and 48 hours, the levels of IL-6 and IL-8 were significantly declined after injecting dexamethasone at the same time(t=0.04,0.01,0.02,0.02, 0.01, 0.01 ,P〈0.05). Conclusion Dexamethasone could protect organism after brain injury by inhibiting the levels of IL- 6 and IL-8.
