摘 要:NR5A2是孤儿核受体家族中的重要成员,在胚胎发育、类固醇激素生成、卵泡发育和排卵中发挥重要作用。经克隆和测序获得二花脸猪NR5A2基因编码区序列,采用RT-PCR技术分析其组织表达特征,并采用荧光定量PCR技术检测二花脸猪与杜长大三元杂交猪卵巢组织中NR5A2基因转录水平。结果表明:二花脸猪NR5A2基因编码区序列长1 506 bp,编码蛋白含有DNA结合域、Ftz-F1盒和配体结合域等保守结构域。二花脸猪NR5A2基因在各种组织中广泛表达,特别是在卵巢组织中高表达,且卵巢组织中转录水平显著高于杜长大三元杂交猪(P<0.05)。推测二花脸猪NR5A2基因在卵泡发育和排卵中发挥重要作用,并可能是影响二花脸猪高繁殖力性状形成的候选基因。NR5A2 is an orphan nuclear receptor that plays important roles in embryonic development, steroidogenesis, follicle development and ovulation. In this study, the coding sequence of NR5A2 gene was obtained by cloning and sequencing techniques in Erhualian pigs, tissue distribution patterns of NR5A2 gene were detected by RT-PCR and transcriptional level was detected in ovarian of Erhualian and Duroc x Landraee x Yorkshire pigs by real-time PCR. The results showed that the length of NR5A2 gene coding sequence was 1 506 bp in Erhualian pigs, and encoding protein had conservative structural domains DBD ( DNA-binding domain) , LBD( ligand-binding domain)and Ftz-F1 box,et al. RT-PCR assays revealed that NR5A2 gene of Erhualian pigs widely expressed in all tissues, especially highly expressed in the ovary tissue, which transcriptional level was significantly higher than that of in Durocx LandracexYorkshire pigs( P〈0. 05 ). Collectively these findings indicate that NR5A2 gene of Erhualian pigs, like other mammalian species NR5A2 gene, plays an important role in follicular development and ovulation,which probably is associated with high fecundity of Erhualian pigs.
