抗解脲脲原体单克隆抗体的制备

摘　　要：作者用提纯的解脲脲原体抗原免疫BALB/c小鼠,取其脾细胞,与小鼠骨髓瘤细胞FO融合,获得3株稳定分泌抗解脲脲原体单克隆抗体的杂交瘤细胞株。制备了小鼠腹水抗体,并对杂交瘤细胞株进行了鉴定。用ELISA法测定该抗体腹水效价为1:64000～1:2048000,1F9、2B2抗体的Ig类型为IgG1,5H1为IgG2b。建立的3株能稳定分泌抗解脲脲原体的McAb杂交瘤细胞林,经液氮冻存后复苏,其分泌抗体能力不受影响。McAbs were prepared by tuning FO cells with spleen cells of BALB/c mice immunized with the antigen of ureaplasma urealyticum. Antibodies in hybrid culture supernatant were detected by ELISA, and positive hybrids were cloned by the technique of limtiing dilutions. Numbers of chromosomes of three hybridoma cull lines ( 1 F9, 2B 2, 5 H 1 ) were demonstrated in the range of 86-96. The immunoglobulin class/subclass of McAb 5 H1 was IgG2b and the others belonged to IgG1. ELISA was used for identifying titer of three McAbs. The titers of three McAbs ( 1 F 9, 2B2, 5H1) in the ascites reached 1:128000, 1 = 64000 and 1:2048000, respectively. After repeated frozen storage and revival, the three hybridoma cell lines can steadily produce high titers of McAbs.
