摘 要:目的探讨染尘大鼠早期单核细胞趋化因子-1(MCP-1)和巨噬细胞炎性蛋白-1α(MIP-1α)的变化规律与炎性细胞及矽肺的关系。方法168只Wistar大鼠随机分为对照组、低、中、高剂量组,每组42只,染尘剂量分别为15、30、60mg/ml。一次性气管内注入染尘建立大鼠矽肺模型,每组分6个时间点(第1、3、7、14、21、28天)分别取7只大鼠处死,收集支气管肺泡灌洗液,采用ELISA试剂盒测定MCP-1、MIP-1α浓度。结果不同染尘剂量间MCP-1、MIP-1α的差异均有统计学意义(P<0.01)。在各时间点剂量组与对照组相比较,2种因子的浓度均有不同程度的升高,差异有统计学意义(P<0.05)。总体观察染尘第28天显示,MCP-1浓度在早期高于后期,MIP-1α在第14天出现高峰,并且MIP-1α与MCP-1、TNF-α、IL-1、IL-6、TGF-β均呈正相关(P<0.01)。BALF中巨噬细胞和中性粒细胞与MCP-1均呈正相关(P<0.05)。结论大鼠肺组织中炎性细胞和细胞因子的改变与MCP-1和MIP-1α趋化因子的改变趋势一致,推断在2种趋化因子的作用下,肺组织中炎性细胞和细胞因子发生相应改变。MCP-1和MIP-1α作为矽肺早期检测生物标志物具有可行性。Objective To explore the variation regularity of monocyte chemotactic protein-1(MCP-1)and macrophage inflammatory protein-1a(MIP-1a)in the early stage of silica exposed rat and the association of inflammatory cells with silicosis.Methods 168 Wistar rats were randomly divided into a control group,and three different dose silica groups(15,30,and 60 mg/ml)with 42 rats in each group.The rats in the control group were instilled with normal saline and the others were instilled intratracheally with silica suspension once only.7 rats out of each group were taken and sacrificed at the 1st,3rd,7th,14th,21st,and 28th d after instillation.Bronchia alveolus lavage fluids(BALF)were collected and the levels of cytokine were determined by enzyme-linked immune-sorbent assay(ELISA).Results The differences of MCP-1 and MIP-1a in different dose groups were statistically significant(P0.01).In comparison of the control group with exposed groups at various time periods,the concentrations of MCP-1 and MIP-1a were elevated in some extent.The differences were significant(P0.05).After 28 days observation,the concentration of MCP-1a at early stage was generally higher than that of later stage;and the concentration of MIP-1a at 14th days reached to a peak.MIP-1a was positively correlated with MCP-1,TNF-a,IL-1,IL-6 and TGF-β(P0.01).In BALF the proportion of macrophages and neutrophils was positively correlated with MCP-1(P0.05).Conclusions Results reveal that the change trend of inflammatory cells and cytokines is consistent with the changes of MCP-1 and MIP-1a in rat lung tissue.It suggests that under the action of two kinds of chemokine the inflammatory cells and cytokines in lung tissue would change correspondingly.It is feasible to regard MCP-1 and MIP-1a as the biomarkers for early detection of silicosis.
