摘  要:目的探讨脂多糖(LPS)对C57BL/6J小鼠海马IL-1β、IL-6、TNF-α及淀粉样蛋白β(Aβ)的影响。方法 50只雄性C57BL/6J小鼠随机均分为五组,分别腹腔连续注射LPS 250μg/kg和生理盐水3d(L3组和C3组)和7d(L7组和C7组);N组作为空白对照组。末次注射后第4天将小鼠进行Morris水迷宫实验;随后处死,测定海马组织促炎细胞因子。结果与C3和C7组比较,L3和L7组小鼠登台潜伏期显著延长,靶象限活动时间显著缩短,海马IL-1β、IL-6及TNF-α表达显著上调(P<0.05)。与L3组和C7组比较,L7组小鼠海马Aβ表达显著上调(P<0.05)。结论 LPS可致小鼠认知功能障碍,可能与海马组织中促炎因子和Aβ表达上调有关。Objeetive To investigate the effects of lipopolysaceharide (LPS) or, cognitive function and proinflammatory cytokines and β-amyloid(Aβ) of the hippocampus in C57BL/6J mouse, Methods Fifty male E57BL/6J mice were equally randomized into 5 groups. LPS 250 μg/kg or normal saline were intraperitoneally injected for 3 consecutive days (group L3 and group C3) and for 7 consecutive days(group L7 and group C7). Group N was taken as the blank control, On the 4th day after the last injection, the mice were tested in the Morris water maze,which then were sacrificed and the expressions of interleukin(IL)-1β, IL-6, tumor necrosis factor(TNF)-α and Aβ in the htppocampus were detected, Results Compared with groups of C3 and C7, the latency to the platform was prolonged, the time spent in the target quadrant was shortened, and the expressions of IL-1β, IL-6, and (TNF)-α in the hippocampus were higher in groups of L3 and LT(P〈0. 05). Compared with groups of C3 and L3, the expression of Aβ in the hippocampus was higher in group L7 (P〈0. 05). Conclusion LPS induces cognitive dysfunction in C57BL/6J mouse, which is probably associated with an upregulation of proinflammatory cytokines and Aβ in the hippocampus.
