摘 要:目的探讨金雀异黄酮对胃癌细胞SGC7901生长的作用及可能机制。方法收集16例胃癌及其相应的癌旁正常组织,通过实时定量PCR分别检测miR-27a及其靶基因ZBTB10的表达水平。用金雀异黄酮0、25、50、100、200μmol/L处理胃癌细胞SGC7901后,采用磺酰罗丹明B染色法检测细胞的生长情况,实时定量PCR检测miR-27a及ZBTB10表达。结果与癌旁正常组织相比,胃癌组织中miR-27a表达增加(P<0.05),而ZBTB10表达降低(P<0.01)。金雀异黄酮可呈浓度、时间依赖的方式抑制胃癌细胞SGC7901的生长,下调miR-27a表达,同时上调ZBTB10表达。结论金雀异黄酮抑制胃癌细胞的生长。其机制可能与下调癌基因miR-27a的表达有关。Objective To investigate the effect and underlying mechanism of genistein on the growth of gastric cancer SGC7901 cells. Methods The mRNA expressions of miR-27a and its target gene ZBTB10 were detected by RT-PCR in 16 cases of gastric cancer and tumor-adjacent normal tissues. After SGC7901 cells were treated with genistein 0,25,50,100,200 μmol/L, cell growth was determined by sulforhodamine'B staining, and the expressions of miR-27a and ZBTB10 were measured by RT-PCIL Results Compared with tumor-adjacent normal tissues, miR-27a expression was increased(P〈0. 05), while ZBTB10 expression was significantly decreased, in gastric cancer tissues (P〈0. 01). Genistein could markedly inhibit the growth of SGC7901 cells in the time- and dose- dependent manners, downregulate miR-27a expression and upregulate ZBTB10 expression. Conclusion Genistein inhibits gi'owth of gastric cancer cells probably via downregulating miR-27a expression.
