慢性炎症诱导的肝脏脂质沉积小鼠模型中抵抗素表达的变化

摘　　要：目的:探讨慢性低度炎症诱导肝脏脂质沉积过程中抵抗素表达和分泌的变化。方法:采用酪蛋白皮下注射18周建立慢性炎症小鼠模型,油红O染色及甘油三酯定量检测肝脏脂质沉积情况,酶联免疫及液态芯片法检测血清炎症因子、抵抗素、游离脂肪酸水平,同时荧光定量PCR检测肝脏、脂肪组织抵抗素和肝脏脂代谢相关基因的表达。结果:成功构建小鼠慢性低度炎症模型,并观察到炎症小鼠血清游离脂肪酸水平增加(P=0.012、P=0.125),胰岛素敏感性降低(P<0.05),肝脏表现为明显的脂代谢紊乱及脂质沉积增加(P<0.05),脂肪组织脂滴变小。同时,炎症小鼠血清抵抗素水平增加(P=0.008、P=0.001),肝脏及脂肪组织抵抗素mRNA表达增加(P<0.05)。结论:慢性炎症诱导的肝脏脂质沉积的小鼠模型中,血清抵抗素水平升高,肝脏及脂肪组织抵抗素mRNA表达增加,提示抵抗素可能在炎症引起的肝脏脂质沉积过程中发挥重要作用。Objective：To examine the expression and secretion of resistin in mice with hepatic lipid deposition induced by chronic inflammation.Methods：Casein was injected into C57BL/6J mice for 18 weeks to induce chronic systemic inflammatory stress in vivo.Serum was analyzed for inflammatory cytokines,resistin,insulin and free fatty acid by ELISA and liquid chip method.Liver was taken for oil red O staining and lipid analysis.Insulin sensitivity was evaluated by insulin tolerance tests.Real-time polymerase chain reaction was used to examine the gene expression of resistin and molecules involved in lipid metabolism.Results：Chronic inflammation mice model was successfully constructed.Casein injection elevated serum levels of free fatty acid in mice（P=0.012,P=0.125）,decreased insulin sensitivity（P0.05）,induced lipid metabolism disorder in the liver,increased lipid deposition（P0.05） and decreased lipid drops of adipose tissues.Meanwhile,serum resistin levels（P=0.008,P=0.001） and mRNA expressions of resistin in the liver and adipose tissues in mice with inflammation were increased（P0.05）.Conclusions：Serum resistin levels and mRNA expressions of resistin in the liver and adipose tissues are elevated in mice with hepatic lipid deposition induced by chronic inflammation,suggesting that resistin may play an important role in liver lipid deposition induced by chronic inflammation.
