摘 要:目的:探讨脾阴虚状态下模型大鼠体内自由基损伤程度和体内抗氧化酶活性的变化,了解脾阴虚在中医脾病发展过程中的地位,为中医脾阴虚证提供现代科学依据,为中医延缓衰老提出新见解。方法 :首先采用"饮食不节+疲劳过度"复合方法造模14 d后,继而给大鼠以温热伤阴药灌胃10 d复制脾阴虚大鼠模型,取大鼠最后24 h尿样测定8-OHdG的含量,取脑皮质和血清测定SOD、GSH-Px、T-AOC、MDA的含量。结果 :脾阴虚模型组大鼠血清和脑皮质中SOD活性、GSH-Px活性、T-AOC和MDA含量与正常组相比均有显著差异(P<0.05)。模型组尿样中8-OHdG含量也有明显上调趋势(P<0.05)。结论 :脾阴虚模型大鼠体内存在着明显的自由基攻击和氧化损伤,但脾阴虚状态处于脾病全程的何种阶段尚需结合前期脾气虚实验数据,并做进一步探究深入了解。Objective : This research was to explore the changing in the spleen yin deficiency rats' which were attacked by free radicals and activity of antioxidant enzymes, understanding the spleen yin deficiency in the developmental process of the spleen deficiency, and providing science evidence for further research. Methods : Fatty diet + over fatigue method was used to establish the model for 14 days, then the rats were given Shangyin drugs for 10 days and the last 24 h urine samples from rats were weighed to measure the content of 8-OHdG, and the SOD, GSH-Px, T-AOC, MDA contents in brain cortex and serum were measured. Results : In serum and cerebral cortex, the contents of SOD, GSH-Px, T-AOC and MDA in the model group showed a significant difference ( P〈0.05 ) .The 8-OHdG in the urine samples of the model group was increased significantly ( P〈0.05 ). Conclusion : There is a clear damage of oxidative free radical in spleen yin deficiency model, but the spleen yin deficiency state in what stage of the spleen disease is needed to understand in-depth studies and combined with previous data of spleen qi deficiency.
