摘 要:目的:探讨生态环境和遗传背景相同条件下3种不同化学变异类型大黄在基因表达水平上的差异,揭示大黄种内变异的分子机制。方法:采用cDNA-SRAP生物信息学方法,对不同化学变异类型大黄地下组织特异表达的基因进行分析和功能预测。结果:共获得5条差异表达基因片段和4条表达量有明显差异的公共条带,其中4条功能已知,分别与植物生长发育及光信号转导、植物抗病性及水分运输相关,5条功能未知。结论:克隆了不同化学变异类型大黄特异表达的基因,为筛选优良变异类型、品种选育、功能基因组的研究提供参考。Objective: To understand the molecular mechanism in genetic variation within species, and identify the differentially expressed genes among 3 anomalies of rhubarb under the same genetic background and cultivated environment. Method: The differentially expressed genes of different anomalies of rhubarb in vigorous growing stage were identified by cDNA-SRAP.Their functions were inferred through bioinformatics method. Result: Five different gene fragments and 4 common gene fragments but differentially expressed were identified, among which, the function of 5 were unknown, and the rest were involved in plant growth, light signal transmitting, disease-resistance, and water transporting. Conclusion: For the first time, the differentially expressed genes were cloned from the different anomalies of rhubarb. This provided the basis of the screening of fine anomalies and varieties, and the research of functional genomics.
