摘 要:目的:建立了茅苍术挥发油中7个成分的体内分析方法。方法:采用内标法测定了给药后不同时间点7个化合物的血药浓度,并通过DAS 3.0软件计算出药代动力学参数,以自定义各成分的权重系数进行茅苍术挥发油整合药代动力学研究,分别获得化合物1~7及其整合的血浆浓度-时间曲线,化合物2~7和整合的药动学参数。结果:整合药动学参数T1/2,Tmax,Cmax,AUC(0-∞)和MRT(0-∞)分别为8.01,4.00 h,3.84 mg.L-1,42.31 mg.L-1.h-1,11.65 h。结论:研究思路符合中医药作用特点,对多组分苍术挥发油的药代动力学做了一些初步的探索。Objective: To investigate pharmacokinetics process of the major components of Atractylodes lancea oil in rats. Method: A preliminary HPLC method based on internal standard method was employed to analysis 7 constituents simultaneously in plasma and to study their pharmacokinetics after oral administration of volatile oil from A. lancea in rats. Pharmacokinetic parameters of compound 2-7 were acquired by DAS 3.0 software and the integrative concentration-time curve was obtained based on the corresponding weighting coefficients. Result: Integrative pharmacokinetic parameters was as follows: T1/2 was 8.01 h,Tmax was 4.00 h,Cmax was 3.84 mg·L^-1,AUC(0-∞) was 42.31 mg·L^-1·h^-1 and MRT(0-∞) was 11.65 h, respectively. Conclusion: This study was thought as an appropriate method for traditional Chinese medicine and we carried out some exploratory survey for pharmacokinetics of volatile oil from A. lancea.
