摘 要:目的:观察不同浓度二苯乙烯苷(TSG)对β-淀粉样肽(Aβ25-35,25μmol.L-1)致损伤的神经干细胞(NSCs)生长、增殖及自我更新能力的影响,探讨TSG对Aβ25-35致NSCs损伤的保护作用。方法:从孕14 d昆明小鼠体外提取神经干细胞,以25μmol.L-1浓度Aβ25-35诱导NSCs建立稳定的AD细胞模型,与不同浓度二苯乙烯苷共同孵育,分为对照组、模型组(Aβ25-3525μmol.L-1)、TSG 1×10-8mol.L-1+Aβ25-35组、TSG 1×10-7mol.L-1+Aβ25-35组、TSG 1×10-6mol.L-1+Aβ25-35组,共5组,分别采用EDU化学荧光染色法检测NSCs增殖情况;采用WST-1细胞增殖及细胞毒性检测法检测不同时段NSCs的细胞活性;体外培养7 d后通过神经干细胞球计数法观察NSCs的自我更新能力。结果:与模型组相比,二苯乙烯苷各剂量组均能在一定程度上抑制Aβ25-35对NSCs的损伤,细胞生长状态良好,细胞增殖率、细胞活性及神经干细胞球计数与模型组相比均有显著性差异(P<0.01)。结论:二苯乙烯苷对Aβ25-35诱导的神经干细胞损伤具有一定保护作用。Objective: To observe the effects of different concentrations of tetrahydroxy stilbene glycoside (TSG) on the growth and proliferation of neural stem cells(NSCs) which were injured by Aβ25-35, to discuss the protective effects of TSG on the NSCs injury induced by Aβ25-35. Method: NSCs were extracted on both sides of brain cortex from the embryos of 14 days of Kunming mice pregnancy,NSCs were cultured by Aβ25-35(25 μmol·L^-1 to establish cell model of Alzheimer's disease (AD) and then were cultured with TSG. All cells were divided into 5 groups,control group; model group(Aβ25-35 25 μmol·L^-1, (TSG 1×10^-8mol·L^-1Aβ25-35)group, (TSG 1×10^-7 mol·L^-1Aβ25-35)group and (TSG 1×10^-6mol·L^-1Aβ25-35)group.Using the chemical fluorescence staining method (EDU) to detect the proliferation of NSCs; applicating WST-1 cell proliferation and cell toxicity test to detect the activity of NSCs of different time and then after cultured 7 days in vitro, counting neurospheres was used to observe the update ability of NSCs. Result: TSG could protect NSCs from Aβ25-35 damage. Compared with model group, the morphology, reproduction rate, cell viability and ball count of NSCs were significant different from TSG group (P〈0.05或P〈0.01). Conclusion: TSG can protect the NSCs injury induced by Aβ25-35 in some ways.
