摘 要:目的研究偏食高碘对大鼠下腔静脉肝段内皮细胞活化的影响。方法将50只断乳后约1个月的远交封闭群SPF/VAF级Wistar大鼠随机分为5组,分别为正常对照(NC)组[常规饲料(碘含量为300μg/kg),饮自来水(碘含量为5μg/L)]、偏食对照(MC)组[偏食饲料(碘含量为300μg/kg),饮自来水]、偏食+10倍碘(M10HI)组(偏食饲料,饮1 850μg/L碘溶液)、偏食+50倍碘(M50HI)组(偏食饲料,饮10 050μg/L碘溶液)、偏食+100倍碘(M100HI)组(偏食饲料,饮20 300μg/L碘溶液),每组10只,雌雄各半。采用自由饮水、摄食的方式进行染毒,连续染毒6个月。采用固相夹心酶联免疫吸附法(ELISA)测定大鼠血浆中细胞间黏附因子-1(ICAM-1)、血管性血友病因子(vWF)及血管性血友病因子前肽(vWFpp)含量。结果与正常对照组相比,无论雄性大鼠还是雌性大鼠,M50HI组和M100HI组大鼠血浆ICAM-1含量及M10HI组、M50HI组和M100HI组大鼠血浆vWF含量以及M50HI组大鼠血浆vWFpp含量均升高,差异有统计学意义(P<0.05,P<0.01);且随着摄入碘含量的逐渐升高,大鼠血浆ICAM-1、vWF含量均呈逐渐上升的趋势。结论高碘可导致偏食大鼠内皮细胞活化,且偏食引起的营养不良可加重高碘所导致的下腔静脉内皮细胞的活化。Objective To observe the effect of monophagia and excessive iodine on the activation of endothelial cells of inferior vena cava in rats. Methods A total of 50 Wistar rats, about one month after delactation, were randomly divided into five groups:normal diet control group (NC)[normal feed (iodine content: 300 μg/kg),tap water (iodine content: 5 μg/L)], monophagia control group (MC)[monophagia feed (iodine content :300 μg/kg),tap water], 10-fold iodine+monophagia group (M10HI) [monophagia feed,iodine solution (iodine content: 1 850 μg/L),50-fold iodine +monophagia group (M50HI) [monophagia feed,iodine solution (iodine content :10 050μg/L), 100-fold iodine+monophagia group (MlOOHI)[monophagia feed, iodine solution (iodine content: 20 300 μg/L), with five males and five females in each group. The rats were treated with iodine through food and drinking water. After six months of exposure,venous blood was collected and ICAM-1 ,vWF,vWFpp were determined. Results Compared with normal control group,no matter male rats or female rats,ICAM-1 increased in M50HI group and MIOOHI group; vWF increased in MIOHI group,M50HI group and M10OHI group and vWFpp in M50HI group increased (P〈0.05, P〈0.01). As the iodine intake increased, ICAM-1, vWF gradually increased. Conclusion Excessive iodine may induce endothelial cell activation in monophagia rats and monophagia diet induced malnutrition can strengthen the effect of high iodine on endothelial cell activation in inferior vena cava of rats.
