摘  要:本试验以Hypodermin C(HC)基因原核表达载体pETHC为模板,PCR扩增HC基因,克隆测序后,亚克隆到真核表达载体pcDNA3.1(+)中,构建了DNA疫苗pcDNA-HC。pcDNA-HC体外转染小鼠胎儿成纤维细胞后,间接免疫荧光技术检测目的基因在转染细胞中的表达。以DNA疫苗pcDNA-HC免疫小鼠,对小鼠进行体液免疫水平检测。结果表明,所构建的DNA疫苗能转染到小鼠胎儿成纤维细胞中并正确表达,免疫小鼠血清IgG滴度随免疫次数的增加而升高。The Hypodermin C(HC) gene of H. Lineatum was amplificated by PCR using pETHC as the tem-plate, cloned and sequenced, then subcloned into pcDNA3.1 (+). AS a result, the pcDNA-HC was successfully constructed. The pcDNA-HC was transfected into mouse fetal fibroblast cells in vitro, and the expression of target gene in transfected cells was detected by indirect immunofluorescence. The results showed that the pcDNA-HC was transfected into mouse fibroblast cells and HC gene was successfully expressed. The IgG titers in immunized mice sera showed a gradual upward trend with the increasing of the immunized times.
