摘  要:本试验旨在阐明湖南省鸡蛔虫分离株线粒体细胞色素C氧化酶第Ⅱ亚基(cox2)基因部分序列(pcox2)和烟酰胺腺嘌呤二核苷酸(NADH)脱氢酶亚单位IV基因(nad4)部分序列(pnad4)的遗传变异情况。利用聚合酶链式反应(PCR)扩增鸡蛔虫的pcox2and pnad4,应用clustalx1.81程序对序列进行比对,结果显示,所获得的cox2(pcox2)and nad4(pnad4)序列长度一致,为350bp(pcox2)and 400bp(pnad4)。由于鸡蛔虫pcox2and pnad4序列种内相对保守,种间差异较大,故可作为种间遗传变异研究的标记,从而为鸡蛔虫的分类鉴定及进一步的分子流行病学调查提供了基础。To analyze the differences of the cytochrome coxidase subunit 2 (pcox2) gene and NADH dehydro-genase subunit 4 (pnad4) gene of Ascaridia galli strains from Hunan province partial sequerces of cox2 (pcox2) and nad4(pnad4) were amplified from each Ascaridia galli sample and were aligned using the Clustal X 1,81. The lengths of all the pcox2 and pnad4 sequences was 350 bp (pcox2) and 400 bp (pnad4). There was no significant variation in pcox2 and pnad4 sequences within Ascaridia galli, while interspecies difference was obvious, SO pcox2 and pnad4 sequences could be used as genetic markers for population genetic studies of roundworms. The results of this study lay down the foundation for further study on molecular epidemiology and diagnostics of Ascaridia galli.
