摘 要:以甘蔗ROC22愈伤组织为材料,研究相关因子对甘蔗胚性细胞团诱导与增殖的效应。结果表明:2,4-D 3.0 mg/L较利于胚性细胞团的诱导,诱导率为30.33%;KT 0.5 mg/L对胚性细胞团的继代增殖较佳,培养25天后的增殖倍数为38.8倍;培养日龄为15天和第4代的胚性细胞团增殖倍数较高,分别为42.06倍和44.33倍;先暗培养再光照培养对胚性细胞团的继代增殖较好,培养25天后的增殖倍数为35.05倍。The callus of sugarcane variety ROC22 was used as the materials to study related factors on the induction and proliferation of embryogenic cell clusters. The results indicated that it was better to induce embryogenic cell clusters with 2,4-D (3.0 mg/L), and the induction rate was 30.33%. It was better to subculture embryogenic cell clusters with KT (0.5 mg/L), and the proliferation times was 38.8 after culture for 25 days. The proliferation time was higher when the age of embryogenic cell clusters was 15 days or 4th successive transfer, which was 42.06 times and 44.33 times respectively. It was better to subculture embryogenic cell clusters when first dark cultivation to illumination cultivation, and the proliferation time was 35.05 after culture for 25 days.
