摘　　要：目的研究miRNA-27a抑制剂antagomiRNA-27a对胶质瘤细胞的影响。方法通过生物信息数据库筛选miRNA-27a预测靶点FOXO3a,并通过基因报告验证预测靶点基因;应用qRT-PCR法检测转染后细胞,MTT法检测antagomiRNA-27a对胶质瘤细胞增殖的影响。结果转染antagomiRNA-27a组细胞增殖受到明显抑制(P<0.05);通过Western blot和报告基因证实FOXO3a是miNAR-27a新的靶基因。结论 miRNA-27a抑制剂对胶质瘤细胞增殖具有抑制作用。Objective To study the effect of antagomiR-27a on cell proliferationin of glioblastoma Methods The predicted target gene of miR-27a was screened out through bioinformatics databases, and the predicted gene was verified using genetic report assay. The effect of antagomiR-27a on the invation and proliferation of glioma cells was analyzed using MTT assay. Results The miR-27a significantly suppressed the invasion of glioblastoma cells（P〈0. 05）. FOXO3a was verified as a new target gene of miR-27a by Western blot and reporter analyzes. Conclusion MiRNA-27a has an inhibitory effect on the proliferation ability of glioblastoma cells.
