摘  要:目的:研究孤核受体鸡卵清蛋白上游启动子转录因子Ⅱ(chicken ovalbumin upstream promoter-tran-scription factor Ⅱ,COUP-TFⅡ)免疫阳性神经元在成年小鼠嗅球中的分布,以及在嗅球内中间神经元中的表达。方法:以Dlx5/6-CIE;COUP-TFⅡflox/+成年小鼠作为研究对象,利用免疫荧光染色方法,结合激光共聚焦扫描技术,观察COUP-TFⅡ免疫阳性神经元在成年小鼠嗅球中的分布。结果:COUP-TFⅡ免疫阳性神经元在Dlx5/6-CIE;COUP-TFⅡflox/+成年小鼠嗅球中广泛表达,主要分布在球周层和颗粒细胞层,在外丛状层中也有少量表达;其中位于颗粒细胞层中的COUP-TFⅡ表达转录因子Sp8,而位于外丛状层和球周层的COUP-TFⅡ则主要分布在Reelin免疫阳性神经元内,COUP-TFⅡ并不在CR+、PV+、Som+、VIP+、CB+和TH+等嗅球中间神经元中表达。结论:成年小鼠嗅球COUP-TFⅡ在Reelin免疫阳性神经元中的表达可能参与嗅球中神经元的迁移和分层。Objective: To investigate the distribution of orphan nuclear receptor chicken ovalbumin upstream promotertranscription factor Ⅱ (COUP-TFⅡ) immunoreactive neurons in the olfactory bulb and the expression of COUP-TFII in the olfactory bulb interneurons. Methods: Adult DlxS/6-CIE; COUP-TFⅡ^flox/+ mice were used as the research objects. Immunofluorescence method combined with confocal laser scanning microscopy was used to investigate the distribution of COUP-TFⅡ immunoreactive neurons in the olfactory bulb of adult mice. Results: COUP-TFⅡ immunopositive neurons were widely distributed in the olfactory bulb of adult Dlx5/6-CIE; COUP-TFⅡ^flox/+ mice, especially in glomerular and granular cell layers, and scattered in external plexiform layer. Moreover, COUP-TFIIⅡ immunopositive neurons located in the granular cell layer express the transcription factor SpS. COUP-TFⅡ were found to be expressed in Reelin immunoposirive interneurons located in glomerular and external plexiform layers, but the expression were not found in the CR + , PV + , Som + , VIP + , CB + or TH + interneurons. Conclusion: COUP-TFⅡ expressed in Reelin interneurons in olfactory bulb might be involved in regulating the migration and layer formation of olfactory bulb interneurons.
