摘 要:利用微卫星标记分析阿根廷滑柔鱼(Illex argentinus)冬生群体与秋生群体的遗传变异性,并检验冬生群体在时间上的遗传差异。结果表明,7个位点平均等位基因数为23.14,平均有效等位基因数为10.43,各位点均为高度多态性位点(PIC=0.590~0.931)。3个群体显示出较高的遗传多样性水平(Ho=0.796~0.904)。两两群体间的Fst值以及AMOVA分析表明,群体遗传分化不显著(Fst=0.008 3,P>0.05);阿根廷滑柔鱼冬生与秋生2个群体间及冬生群体在时间上的遗传差异不显著。基于Nei’s遗传距离构建的UPGMA聚类树显示,冬生群体2个群体聚在一起。阿根廷滑柔鱼为大型洄游性种类,不同产卵群体有可能在生殖洄游过程中发生混合并进行基因交流,从而表现出遗传同质性。Genetic differentiation between winter spawning cohort and autumn spawning cohort of Illex argentinus was evaluated and the impact of temporal variation on winter spawning cohort at one location was measured using 7 microsatellite DNA loci. The results showed that the number of average alleles and average effective alleles was 23.14 and 10.43 respectively. All microsatellite markers were highly polymorphic (PIC = 0. 590 -0. 931 ) and the three groups contained high genetic variations (Ho = 0. 796 -0. 904). The pairwise fixation index F-statistics (Fst) and analysis of molecular variance (AMOVA) showed that nosignificant genetic differentiation existed between winter spawning cohort and autumn spawning cohort and no significant effect on genetic diversity of the temporal variation in winter spawning cohort was detected. UPGMA clustering tree based on Nei' s genetic distance demonstrated that two groups of winter spawning cohort clustered together. Being a migratory species, different spawning cohorts of I. argentinus may mingle during their spawning migration, and the frequent gene flows may occur, which makes two spawning cohorts genetically homogeneous.
