妊娠期高血压疾病患者早期血浆蛋白质的表达

摘　　要：目的:探讨炎症状态和妊娠期高血压疾病(HDCP)发生的关系,初步筛选HDCP早期标志物。方法:采用病例对照的方法研究601例HDCP患者和1 053例正常妊娠妇女血液系统白细胞参数,分析其在两组中的变化趋势;采用蛋白芯片筛选507种蛋白在1∶1匹配的8例妊娠20周前孕妇血浆中的表达情况。结果:妊娠20周前,HDCP患者的白细胞参数比对照组明显升高,两组中有5类31种蛋白表达水平差异有统计学意义:除GH升高外,血管生长因子(血管生成素样蛋白2、EG-VEGF和PIGF)、炎症反应因子(IL-1 sRⅡ、TWEAK、IFN-α/β、IL-2、IL-6 R、MIP-3β、RANK、CCR6、TLR4、OX40配体、趋化因子、白细胞衍生趋化因子2、TNF-α)、生长因子(IGF-ⅡR、IGFBP-1、FGF-21、血小板反应蛋白-4、GM-CSF)、基质金属蛋白(MMP-11、MMP-1、MMP-10)和其他蛋白(食欲素B、TMEFF2、卷曲蛋白-6、XEDAR、抑制素-A和泛激素)表达降低。结论:证实炎症与HDCP发生有关,筛选出的31种蛋白可能是HDCP潜在的早期标志物。Objective： To explore the relationship between inflammation condition and the development of hypertensive disorders complicating pregnancy （HDCP） , and screen the early markers of HDCP. Methods： The blood system leukocyte parameters of 601 HDCP patients and 1 053 normal pregnancies as control were studied and the change trend of the two groups was analyzed. The expression of 507 proteins were examined by antibody array technology. Results ： Women with HDCP presented significantly higher leukocyte parameters in the second trimester compared to the normal gravidas. 5 series 31 kinds proteins expressed differently between the two groups. Except GH increasing, vascular growth factors （ angiopoietin - like 2, EG - VEGF and PIGF） , inflammation factors （ IL - 1 sR II , TWEAK, IFN - alpha/beta, IL-2, IL-6 R, MIP-3 beta, RANK, CCR6, TLR4, OX40 ligand, chemotatic factor, leucocyte chemotatic factor-2, TNF- alpha）, growth factor （IGF- I] R, IGFBP- 1, FGF- 21, platelet reaction protein -4, GM -CSF）, matrix metalloproteinases （MMP-11, MMP- 1, MMP- 10） and other proteins （orexin B, TMEFF2, frizzled protein-6, XEDAR, inhibin- A and ubiquitin） were in lower expression. Conclusion： TheRE is a relationship between inflammation and HDCP occurring, selected 31 proteins may be potential inflammatory markers to HDCP.
