九味头痛片预给药对偏头痛大鼠软脑膜血流量和脑组织5-羟色胺、一氧化氮水平的影响  

Effect of Jiuwei Headache Tablet on Cerebral Blood Flow,5-HT and NO of Brain Tissues in Rats Migraine Model

摘　　要：目的:观察九味头痛片(JWTTP)预给药对偏头痛大鼠软脑膜血流量、脑组织一氧化氮(NO)水平、5-羟色氨(5-HT)的调节作用。方法:将实验大鼠分为正常对照组、偏头痛模型组、正天丸治疗组、JWTTP高、中、低剂量治疗组共6组,制模前连续预给药7天(正常对照组、偏头痛模型组除外),于末次给药后30min,除正常对照组外,每只大鼠按10mg/Kg体重皮下注射硝酸甘油,复制偏头痛动物模型。用激光多普勒血流仪检测各组大鼠软脑膜血流量,用化学一步法及免疫组织化学法分别检测各组大鼠脑组织NO水平和5-HT阳性细胞率。结果:与正常对照组比较,偏头痛大鼠血管调节功能紊乱、软脑膜血流量降低,NO水平升高,5-HT阳性细胞率也较高(P<0.01)。JWTTP高、中、低剂量组与模型组比较,软脑膜血流量均明显增加(P<0.01);脑组织NO水平均显著降低(P<0.05或P<0.01);5-HT阳性细胞率均明显减少(P<0.05或P<0.01);JWTTP高剂量组治疗效果优于正天丸治疗组。结论:JWTTP可能通过调节神经递质,改善脑组织血供,有效调理血管舒缩紊乱而发挥对偏头痛的治疗作用。Objective: To study the mechanism of Jiuwei headache tablet(JWTTP)on migraine rats model through observing the changes in the cerebral blood flow ( CBF ), thrombocytin(5-HT) and nitrogen monoxidum(NO) levels in brain tissues. Method: The rats were randomly divided to six groups:control,model,Zheng-Tian Wan(ZTW), high, medium and low dose of JWTTP groups. The drugs were administrated continuously before modeling besides control and model groups. The model was established by injecting nitroglycerin (10mg/Kg) subcutaneous except control group at 0.5h after the last drug administration. The cerebral blood flow was detected by Laser Doppler probe(407), NO levels and 5-HT positive cells rates in rats brain tissues were determinated by biochemical and immunohistochemical methods respectively. Results: Compared with model group, the cerebral blood flow was significantly increased in high, medium and low dose of JWTTP groups(P<0.01);NO levels was significantly decreased (P<0.05, P<0.01);5-HT positive cells number and positive cells rates was significantly decreased(P<0.05, P<0.01). Conclusion: JWTTP has a therapeutical effect on migrain through improving cerbral blood flow, regulating neurotransmitter levels and the constriction or dilation of blood vessels.
