摘 要:采用改良的CTAB法提取万寿菊(Tagetes erecta L.)叶斑病病原菌总DNA,以ITS1和ITS4为引物扩增病原菌的ITS片段,PCR扩增产物纯化后测序并与数据库中的已知序列进行BLAST比对,构建分子系统发育树,对该病原菌进行分子鉴定。结果显示分离的万寿菊叶斑病病原菌ITS序列与GenBank中万寿菊链格孢(Alternaria tagetica)AF267134的ITS序列相似度为99%,在分子系统发育树上与万寿菊链格孢菌株聚为一支,结合形态特征确定分离的万寿菊叶斑病病原菌为万寿菊链格孢。Total DNA of pathogenic fungus of marigold(Tagetes erecta L.) leaf spot was extracted by modified CTAB method. The ITS segment was amplified using ITS1 and ITSg as primers, then purified and sequenced. The ITS sequence obtained was blasted with known ITS sequences in database, and the phylogenic tree was constructed. The results showed that the ITS sequence of the fungus had 99% similarity with the ITS of Alternaria tagetica AF267134 in GenBank. The phylogenic tree al- so showed that the pathogenic fungus and A. tagetica clustered on the same branch. Combining with morphological characteris- tics, the pathogenic fungus of marigold leaf spot was identified as Alternaria tagetica.
