摘 要:采用改良的CTAB法、SDS法和试剂盒法提取柽柳(Tamarix chinensis Lour)和美人蕉(Canna indicaLinn)不同组织中的RNA,通过紫外分光光度法和琼脂糖凝胶电泳对提取的RNA质量进行检测。结果表明,CTAB法从柽柳和美人蕉叶中提取的RNA A260 nm/A280 nm分别为1.96和1.91,纯度较高。琼脂糖凝胶电泳结果显示CTAB法和SDS法提取的植物RNA 28 S rRNA和18 S rRNA条带清晰明亮,说明提取的RNA完整性好,浓度较高;而试剂盒法提取的植物RNA存在降解现象和杂质干扰。Modified CTAB method, SDS method and kit method were applied to extract RNA from different tissues of Tamar-ix chinensis Lour and Canna indica Linn, the quality of extracted RNA was detected by UV spectrophotometry and agarose gel electrophoresis. The results showed that the A260nm/A280nm of RNA extracted from leaves of T. chinensis and C. indica by CTAB method was 1.96 and 1,91, respectively, indicating high purity of RNA. Agarose gel electrophoresis showed that RNA of the two plants extracted by CTAB and SDS method was with clear and bright 28 S rRNA and 18 S rRNA bands, indicat- ing the integrality and concentrating of RNA was well; while the RNA extracted by kit method was degraded and with impu- rity interference.
