摘  要:目的:研究高脂高蛋白饮食对吡非尼酮药动学的影响。方法:10名健康受试者空腹单次口服吡非尼酮片400mg,经1周清洗期,给予高脂高蛋白饮食同时单次口服吡非尼酮片400mg。采用高效液相色谱法测定血药浓度,用SPSS13.0统计软件计算两者的药动学参数,并评价高脂高蛋白饮食对吡非尼酮药动学的影响。结果:空腹组和高脂高蛋白饮食组主要药动学参数分别为:tmax(0.63±0.23)、(0.70±0.19)h,cmax(11.97±1.34)、(9.21±1.24)μg/ml,t1/2(0.33±0.10)、(0.33±0.09)h,AUC0-12h(29.54±8.50)、(25.20±7.77)μg.h/ml,其中cmax比较差异有统计学意义(P<0.001)。结论:高脂高蛋白饮食可影响吡非尼酮的药动学参数。OBJECTIVE: To study the effect of high lipid and protein diet on the pharmacokinetics of pirfenidone. METHODS: 10 healthy volunteers received single oral dose of Pirfenidune tables 400 mg on an empty stomach, or in combined with high lipid and protein diet after a week cleaning period. Plasma concentrations of pirfenidone were measured by HPLC. The pharmacokinetic pa- rameters were calculated by SPSS software (version 13.0), and the effect of high lipid and protein diet on pirfenidone was evaluated. RESULTS: The main pharmacokinetic parameters were as follows in empty stomach group and high lipid and protein diet group: tmax (0.63 ± 0.23)h and (0.70 ±0.19) h; cmax(11.97 ± 1.34) gg/ml and (9.21 ± 1.24) gg/ml; t1/2(0.33 ± 0.10) h and (0.33 ± 0.09) h; AUC0-12 h(29.54 ± 8.50)μg/h. mL and (25.20 ± 7.77) ixg-h/ml, respectively. There was significant difference between the two groups in the cmax(P〈0.001 ). CONCLUSIONS: High lipid and protein diet can influence the laharmacokinetic oarameters of oirfenidone.
