TNF-α、MMP-2及NF-κB在人子宫肌瘤组织中的表达研究

摘　　要：目的:检测肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-2(MMP-2)及核转录因子(NF-κB)在人子宫肌瘤中的表达,探讨其在子宫肌瘤发病中的作用,为临床治疗提供理论依据。方法:收集30例子宫肌瘤组织(HL)和与之配对的子宫肌组织(HM),进行原代细胞培养;蛋白免疫印迹和实时聚合酶链反应检测TNF-α、MMP-2在HL和HM及原代培养细胞中蛋白及mRNA的表达。结果:(1)卵泡期子宫肌瘤组织中、人子宫肌瘤平滑肌细胞(HL-SMCs)中TNF-α、MMP-2的表达均高于正常子宫肌组织及人子宫平滑肌细胞(HM-SMCs),黄体期两者表达均无差异。(2)NF-κB信号通路抑制剂Bay11-7082能够抑制卵泡期HL-SMCs中TNF-α对MMP-2蛋白的诱导作用。结论:TNF-α、MMP-2在卵泡期子宫肌瘤中均高表达,TNF-α还可通过激活HL-SMCs中NF-κB信号通路,进一步促进该期子宫肌瘤中MMP-2的表达。Objective： To investigate the mechanism and effect of TNF-α, MMP-2 and NF-κB in development of uterine leiomyoma and provide the basis of theory for instructing its clinical treatment. Methods：30 pairs of human uterine leiomyoma tissues （HL） and their matched myometrial tissues （HM） were collected for primary cell culture and protein and mRNA extraction, the expression level of TNF-α,MMP-2 in tissues and in HL-SMCs and primary cultured HM-SMCs were detected by Western blotting and real-time PCR. HL-SMCs and HM- SMCs were treated with TNF-α and Bay 11-7082, a selective inhibitor of NF-κB activity, the expression of MMP-2 was analysised again. Results：（1）Western blots and real-time PCR showed that TNF-α expression in leiomyoma tissue and cells was higher than that in the adjacent normal myometrial tissue and cells during the follicular phase , whereas there was no difference in the luteal phase. （2） The Bay11-7082 only blocked TNF-induced the expression of MMP-2 in HL-SMCs, but not in HM-SMCs during the follicu- lar phase. Conclusion：The study indicates that TNF-α and MMP-2 are increased in human leiomyoma compared with corresponding myometrium during the follicular phase. TNF-α induces the expression of MMP-2 through the activation of NF-κB signaling pathways in HL-SMCs during the follicular phase.
