摘  要:分别以巨尾桉、尾叶桉成熟种子和茎段为组培苗初始材料进行继代培养,利用ISSR标记对不同无性系、不同继代次数(1~30代)的巨尾桉、尾叶桉组培苗进行遗传稳定性检测。结果表明:ISSR分子标记在桉属植物鉴定中具有较高的灵敏度,可以检测到巨尾桉和尾叶桉种间和种内的遗传差异。而对同一无性系不同继代次数的个体进行扩增时则未发现其在带型和带的强弱上的差异,证明组培苗在经过多次继代培养后,在DNA水平上未发生变异,多代培养后的实验材料仍能保持遗传上的稳定性。The genetic stability of Eucalyptus grandis ×E.urophylla and Eucalyptus urophylla,which were repeatedly subcultured in vitro for one to thirty times with two different original materials of mature seed and stem segment,was examined by using ISSR.The results showed that the ISSR molecular markers in eucalyptus had higher sensitivity,which can detected the interspecific and intraspecific genetic differences of Eucalyptus grandis and Eucalyptus urophylla.For the amplification individuals of different subculturing times in the same clone,differences of strong and weak in banding pattern were not found.It explained that tissue culture seedlings DNA level remained the same after many subculturing,the material can still keep the genetic stability after many successive generations cultivation.
