沼泽红假单胞菌的分离鉴定、纯化及计数培养基优化研究

摘  要:目的:通过分离鉴、纯化与鉴定菌株,然后按沼泽红假单胞菌所需的营养组分,优化计数培养基的配方,为生产中的计数提供试验数据.方法:待分离的菌落接种于培养基,然后进行反复的纯化,最后观察菌落的形态,按照细菌鉴定手册鉴定;按所需碳源、氮源、磷酸盐、酵母膏等因素进进行正交设计,优化出最佳的培养基配方.结论:分离与纯化得到一株红红螺菌科沼泽红假单胞菌;通过优化得到最优及验证试验得到最佳的计数培养基配方为:氯化铵1.5 g/L,乙酸钠1.5 g/L磷酸氢二钾0.4g/L,氯化钠1.0g/L,硫酸镁0.3g/L,酵母膏2.0g/L,琼脂14.0g/L.该配方能够对沼泽红假单胞菌准确及简捷的计数.Objective: By separating, purifying and identifying authentication strains, and then optimizing counting medium component according to swamp red pseudomonas with the nutrition component, to provides counting experimental data for production. Method: Separating strain vaccination in medium, then repeatedly purifying, and finally observing the character of the strain according to the bacteria identification manual; conducting orthogonal design according to the carbon source, nitrogen source, phosphate, and yeast extract factors to optimize the medium component. Conclusion: Through separation and purification, a plant of red rhodospirillaceae swamp with red pseudomonas is obtained; Through optimization and verification test, the best counting medium formula is proved to be: ammonium chloride 1.5g, sodium acetate 1.5 g, dipotassium hydrogen phosphate 0.4 g and Lsodium chloride 1.0 g, magnesium sulfate 0.3 g, yeast extract 2.0 g, agarl4g. This medium is accurate and convenient in counting the number of bacterium Rhodopseudomonas palustris.
